摘  要:【目的】探讨长期给药建立稳定高效的高尿酸血症合并肾损伤的大鼠模型。【方法】将48只雄性SD大鼠随机分为正常对照组、氧嗪酸钾组、腺嘌呤组、氧嗪酸钾+腺嘌呤组、酵母膏+氧嗪酸钾组以及酵母膏+腺嘌呤组,每组8只,分别给予5%羧甲基纤维素钠2 mL/d灌胃、氧嗪酸钾250 mg·kg^(-1)·d^(-1)灌胃、腺嘌呤200 mg·kg^(-1)·d^(-1)灌胃、氧嗪酸钾250 mg·kg^(-1)·d^(-1)联合腺嘌呤200 mg·kg^(-1)·d^(-1)灌胃、酵母膏15 g·kg^(-1)·d^(-1)联合氧嗪酸钾250 mg·kg^(-1)·d^(-1)灌胃以及酵母膏15 g·kg^(-1)·d^(-1)联合腺嘌呤200 mg·kg^(-1)·d^(-1)灌胃。连续干预4周后,检测血清尿酸、尿素氮和肌酐水平,采用苏木素-伊红染色法评估肾脏组织病理学变化。【结果】在为期4周的造模后,各造模组血清尿酸、尿素氮和肌酐值均显著高于正常对照组(P <0.05)。其中,氧嗪酸钾+腺嘌呤组效果最明显。各造模组可见肾组织尿酸钠盐结晶体积聚,肾小管周围炎性细胞浸润严重,上皮细胞胞质空泡明显。【结论】为期4周的氧嗪酸钾、腺嘌呤和酵母膏处理均可以建立稳定高效的高尿酸血症的动物模型。Objective To establish stable and efficient rat model of hyperuricemia complicated with renal injury by long-term administration. Methods Forty-eight male SD rats were randomly divided into normal control group,potassium oxonate group,adenine group,potassium oxonate + adenine group,yeast extract + potassium oxonate group and yeast extract + adenine group, with 8 rats in each group. Above groups were given 5% of sodium carboxymethylcellulose 2 m L/d by gavage,potassium oxonate 250 mg·kg^(-1)·d^(-1) by gavage,adenine 200 mg·kg^(-1)·d^(-1) by gavage,potassium oxonate 250 mg·kg^(-1)·d^(-1)combined with adenine 200 mg·kg^(-1)·d^(-1) by gavage,yeast extract 15 g·kg^(-1)·d^(-1)combined with potassium oxonate 250 mg·kg^(-1)·d^(-1) by gavage and yeast extract 15 g·kg^(-1)·d^(-1)combined with adenine 200 mg·kg^(-1)·d^(-1) by gavage,respectively. After 4 weeks of continuous intervention,serum uric acid,urea nitrogen and creatinine were detected,and hematoxylin-eosin staining was used to evaluate the renal pathological changes. Results After 4 weeks of modelling,serum uric acid,urea nitrogen and creatinine values in all modeling groups were significantly higher than those of in the normal control group(P < 0.05). The effect was most pronounced in the potassium oxonate + adenine group. In each model group, there was an accumulation of sodium urate crystals in the renal tissue,with severe peritubular inflammatory cell infiltration and marked cytoplasmic vacuolation of the epithelial cells. Conclusion A 4-week treatment of potassium oxonate,adenine and yeast extract is effective for establishing a stable and efficient animal model of hyperuricaemia.
