摘 要:目的探讨三氧化二砷(As_(2)O_(3))通过早幼粒细胞白血病(PML)基因对三阴性乳腺癌细胞增殖、侵袭迁移的影响。方法采用慢病毒介导的短发夹核糖核酸干扰技术,观察PML对三阴性乳腺癌MDA-MB-231细胞增殖、克隆形成和侵袭迁移的影响,进一步观察As_(2)O_(3)联合沉默PML表达对MDA-MB-231细胞增殖、克隆形成和侵袭迁移的影响。结果沉默PML的表达可以降低MDA-MB-231细胞的增殖活性,减少细胞的克隆数量,抑制细胞的侵袭转移能力。As_(2)O_(3)联合PML沉默可以显著抑制细胞增殖。As_(2)O_(3)联合PML沉默组的细胞克隆数最少,细胞迁移数最少。结论沉默PML的表达可以降低三阴性乳腺癌细胞的增殖、侵袭迁移能力,As_(2)O_(3)可以增强PML对细胞增殖和侵袭迁移能力的抑制作用,可能作为乳腺癌新的治疗靶点,对三阴性乳腺癌具有潜在的治疗价值。Objective To study the effect of arsenic trioxide(As_(2)O_(3))on cell proliferation,invasion and migration of triple-negative breast cancer cells through the promyelocytic leukemia(PML)gene.Methods Lentivirus-mediated short hairpin ribonucleic acid interference technology was used to observe the effects of PML on the proliferation,clone formation,invasion and migration of MDA-MB-231 cells;further observe the effects of arsenic trioxide combined with PML expression silence on breast cancer cell proliferation,clone formation,invasion and migration.Results PML silence in MDA-MB-231 cells inhibited the cell proliferation,reduced the number of clones,and inhibited cell invasion and migration.As_(2)O_(3) treatment combined with PML silence inhibited cell proliferation.The number of cell clone and cell migration in the As_(2)O_(3) combined with PML silence group was the least,and with significant difference compared with PML silence group.Conclusion PML silence reduced the proliferation,invasion and migration ability of triple-negative breast cancer cells,and As_(2)O_(3) enhanced the inhibitory effect of PML on cell proliferation,invasion and migration.As_(2)O_(3) treatment combined with PML silence could be used as a new therapeutic target for breast cancer and has potential therapeutic value for triple-negative breast cancer.
