摘  要:目的阐明Wnt/β-catenin通路对Foxo3a及其下游分子的调控在多囊卵巢综合(PCOS)征患者颗粒细胞中的作用机制。方法选取在接受体外受精-胚胎移植(IVF-ET)或者卵胞浆内单精子显微注射(ICSI)助孕治疗的PCOS患者及单纯输卵管因素患者为研究对象。分离、纯化患者取卵日卵泡液中的卵巢颗粒细胞,采用实时定量PCR方法检测颗粒细胞中β-catenin、Foxo3a、P27、Cyclin D1、Caspase3、Caspase8的表达水平。结果与对照组相比,PCOS患者卵巢颗粒细胞中β-catenin、Foxo3a、P27、Caspase3、Caspase8表达水平升高,P<0.05或<0.01,而两组患者卵巢颗粒细胞中的Cyclin D1表达水平的差异无统计学意义。结论PCOS患者卵巢颗粒细胞的凋亡增加,增殖能力下降。Wnt/β-catenin通路对Foxo3a及其下游信号分子的调控参与了PCOS患者卵巢颗粒细胞异常的机制。Objective To clarify the mechanism of the Wnt/β-catenin pathway regulating Foxo3a and its downstream molecules in the granulosa cells of patients with polycystic ovary syndrome.Methods Patients with polycystic ovary syndrome who received in vitro fertilization-embryo transfer(IVF-ET)or intracytoplasmic sperm injection(ICSI)and patients with simple fallopian tube factors were selected as the research subjects.Ovarian granulosa cells were separated and purified from the follicular fluid of patients on the day of egg retrieval.Quantitative PCR was used to detect the expression levels ofβ-catenin,Foxo3a,P27,Cyclin D1,Caspase8,and Caspase3 in the granulosa cells.Results Compared with those in the control group,the expression levels ofβ-catenin,Foxo3a,P27,Caspase8,and Caspase3 in ovarian granulosa cells of patients with polycystic ovary syndrome increased,and the difference was statistically significant(P<0.05 or P<0.01).However,there was no statistical difference in the expression level of Cyclin D1 in ovarian granulosa cells between the two groups.Conclusion The apoptosis of ovarian granulosa cells in patients with polycystic ovary syndrome increases and the proliferation ability decreases.The regulation of Foxo3a and its downstream signal molecules by Wnt/β-catenin pathway is involved in the abnormal mechanism of ovarian granulosa cells in PCOS patients.
