摘  要:目的研究睑板腺功能障碍(MGD)与非MGD(NMGD)患者的眼表菌群的组成及差异。方法招募了57例MGD患者为MGD组,32名健康志愿者为NMGD组,对两组受试者结膜拭子进行16S rRNA基因V3-V4片段的高通量测序,对测序数据进行生物信息学分析,以研究MGD组患者和NMGD组受试者眼表菌群的组成及差异。结果MGD组患者和NMGD组受试者眼表菌群的16S rRNA的测序结果显示,眼表菌群多样性没有差异(均为P>0.05)。生物信息学分析结果显示,在门和属水平上两组受试者显示了相似的眼表菌群构成。MGD组患者中,I级、II级和III级患者Shannon指数(P=0.006)和Dominance指数(P=0.012)均存在显著差异。此外,使用未加权和加权的Unifrac方法构建的主坐标分析图也表明,I级、II级、III级MGD患者之间的样本区分明显(P=0.039)。结论有无MGD的患者眼表菌群没有显著差异。不同严重程度的MGD患者的眼表菌群存在显著差异。Objective To investigate the composition and difference of ocular surface(OS)microbial colonies between patients with meibomian gland dysfunction(MGD)and non-meibomian gland dysfunction(NMGD).Methods A total of 57 MGD patients were recruited in the MGD group,and 32 healthy volunteers were recruited in the NMGD group.The high-throughput sequencing of 16S rRNA V3-V4 fragments was performed using their conjunctival swab samples.The sequencing data was analyzed bioinformatically to explore the composition of the OS microbial colonies in MGD and NMGD patients and their difference.Results The 16S rRNA sequencing data showed that there was no significant difference in the diversity of OS microbial colonies between the MGD group and the NMGD group(both P>0.05).The bioinformatic analysis results showed there was a similar OS microbial structure at the phylum and genus levels between the two groups.In the MGD group,Grade I,Grade II and Grade III MGD patients showed significant differences in Shannon’s index(P=0.006)and dominance index(P=0.012).Furthermore,the principal coordinate analysis chart constructed using the unweighted and weighted Unifrac method also demonstrated a significant difference among samples of patients with Grade I,Grade II and Grade III MGD(P=0.039).Conclusion There is no significant difference in the OS microbial colonies between MGD patients and NMGD patients.However,there is a significant difference in the OS microbial colonies among patients with different grades of MGD.
