摘  要:为确定从腌韭菜根中获得的土扬芽孢杆菌、巨大芽孢杆菌、莴苣不动杆菌、解淀粉芽孢杆菌、蜡样芽孢杆菌五种腐败菌的菌液OD值与活菌数之间的线性关系,从而建立一种能快速获得菌悬液活菌浓度的方法。采用比浊法测定不同浓度菌液的OD_(600)值,并通过平板计数法测得各菌液活菌数,比较二者之间的相关性,建立标准曲线。结果表明,腌韭菜根中五种腐败菌菌液的活菌数与OD_(600)值呈直线线性相关,其线性方程为:土扬芽孢杆菌:y=25.81x+3.25,R^(2)=0.9842;巨大芽孢杆菌:y=16.63x+2.64,R^(2)=0.9357;莴苣不动杆菌:y=16.16x-0.0035,R^(2)=0.9268;解淀粉芽孢杆菌:y=7.25x+0.30,R^(2)=0.9926;蜡样芽孢杆菌:y=40.31x+0.83,R^(2)=0.9890。该研究结果可为五种腐败菌菌液中活菌数量的测定提供依据。Five kinds of spoilage bacteria were isolated from the pickled Chinese chives roots,including Bacillus toyonensis,Bacillus megaterium,Acinetobacfer lactucae,Paenibacillus amylolyticus and Bacillus cereus.We determined the linear relationship between the OD value and the number of live bacteria of the five kinds of spoilage bacteria,so as to establish a method to quickly obtain the live bacteria concentration of the bacterial suspension.The OD_(600) values of different concentrations of bacterial suspension were determined by turbidimetric method,and the numbers of live bacteria were measured by plate counting method.The correlation between the two was analyzed,and a standard curve was established.The results showed that there was a linear correlation between the number of live bacteria and OD_(600) value,and the linear equations were as follows:Bacillus toyonensis:y=25.81x+3.25,R^(2)=0.9842;Bacillus megaterium:y=16.63x+2.64,R^(2)=0.9357;Acinetobacfer lactucae:y=16.16x-0.0035,R^(2)=0.9268;Paenibacillus amylolyticus:y=7.25x+0.30,R^(2)=0.9926;Bacillus cereus:y=40.31x+0.83,R^(2)=0.9890.This study has provided reference for the determination of the number of live bacteria in bacterial suspension.
