摘 要:目的检测oHSV2(oncolytic herpes simplex virus type 2,oHSV2)所携带的外源基因萤火虫荧光素酶(firefly luciferase,Fluc)能否在BALB/c小鼠皮下表达。方法选取20只雌性BALB/c小鼠,用小鼠结直肠癌细胞(CT26)植瘤10只小鼠,采用瘤内和皮下两种方式给药分别用高剂量和低剂量oHSV2-FLuc,使用小动物活体成像仪观察BALB/c小鼠体内荧光信号表达情况,每隔24h观察一次,持续观察7d。结果BALB/c小鼠经皮下给药后,活体成像系统能够检测出Fluc所发出的荧光信号。结论oHSV2所携带的外源基因可以在皮下正常表达。Objective To determine whether the Firefly Luciferase(FLuc)genes carried by oncolytic herpes simplex virus type 2(oHSV2)could be expressed subcutaneously in BALB/c mice.Methods A total of 20 female BALB/c mice were selected and 10 mice were treated with mouse colorectal cancer cells(CT26)xenograft.High and low doses of oHSV2-Fluc were respectively administered in both intratumoral and subcutaneous ways.The fluorescence signal expression in BALB/c mice was observed by small animal imaging machine every 24h.The observation was continued for 7 days.Results Fluorescence signals from BALB/c mice could be detected by the in vivo imaging system after subcutaneous administration.Conclusion The exogenous genes carried by oHSV2 can be expressed subcutaneously.
