摘 要:目的检测血小板反应蛋白-2(thrombospondin-2,THBS2)在胰腺导管腺癌(pancreatic ductal adenocarcinoma,PDAC)组织及癌旁正常组织中的表达情况,并探讨其表达与临床病理特征及预后的关系。方法采用免疫组化EliVision法检测69例PDAC组织及癌旁正常组织中THBS2的表达,分析THBS2表达与PDAC临床病理特征的关系。利用生物数据库分析PDAC组织中THBS2表达的差异及对患者生存的影响。结果PDAC组织中THBS2阳性率为65.2%(45/69),癌旁正常组织中的阳性率为36.2%(25/69)。THBS2在PDAC组织中的阳性率显著高于癌旁正常组织(P<0.05)。THBS2表达与肿瘤分化程度、临床分期及淋巴结转移密切相关(P<0.05)。在PDAC中,肿瘤分化程度越低、TNM分期越高者,THBS2阳性率越高;THBS2表达与患者性别、年龄、肿瘤位置、肿瘤直径、血管侵犯无关(P>0.05)。基因表达谱交互分析数据库(Gene Expression Profiling Interactive Analysis,GEPIA)显示,THBS2在胰腺癌组织的表达显著高于癌旁正常组织(P<0.01)。Kaplan Meier-plotter在线生存分析软件显示,THBS2高表达患者的总生存期低于低表达患者(HR=1.57,P<0.05)。结论PDAC组织中THBS2高表达,其在肿瘤发生、发展过程中发挥重要作用,且与患者的预后密切相关,有望成为胰腺癌预后评估的重要生物标志物及潜在分子治疗靶点。Purpose To detect the expression of thrombospondin-2(THBS2)in pancreatic ductal adenocarcinoma(PDAC)tissue and adjacent normal tissues,and to explore the relationship between its expression differences and clinicopathological characteristics and prognosis of patients.Methods The immunohistochemical method was used to detect the expression of THBS2 in 69 cases of PDAC and corresponding normal tissues adjacent to the cancer.Statistical methods were used to analyze the expression of THBS2 and the clinicopathological characteristics of PDAC patients.The GEO database was used to analyze the effect of THBS2 expression in PDAC tissue on patients’survival.Results The positive expression rate of THBS2 in PDAC tissues was 65.2%(45/69),and the positive expression rate in the corresponding adjacent tissues was 36.2%(25/69).The positive expression rate of THBS2 was significantly higher than that in adjacent tissues(P<0.05).The expression of THBS2 was closely related to the degree of tumor differentiation,clinical stage and lymph node metastasis(P<0.05).In PDAC,the lower the degree of tumor differentiation and the higher the TNM stage,the higher the positive expression rate of THBS2.There was no significant correlation between the expression of THBS2 and the patient’s gender,age,tumor location,tumor diameter,and vascular invasion(P>0.05).The GEPIA database analysis showed that the expression of THBS2 in pancreatic cancer tissues was significantly higher than that of normal in adjacent tissues(P<0.01).Kaplan-Meier plotter online survival analysis showed that the overall survival of patients with high THBS2 expression was significantly lower than that of patients with low expression(HR=1.57,P<0.05).Conclusion THBS2 is highly expressed in PDAC tissues,plays an important role in the occurrence and development of tumors,and is closely related to the prognosis of patients.It is expected to become an important biomarker and potential molecular therapy target for prognostic evaluation of pancreatic cancer.
