摘  要:目的分析PCDH9在调控前列腺癌细胞周期中的作用,探讨其表达缺失对抑制前列腺癌细胞凋亡、促进肿瘤进展中的分子机制。方法采用免疫组化SP法检测92例前列腺癌和36例良性前列腺组织中PCDH9、STAT3和细胞周期调控基因Cyclin D1和C-myc的表达情况,分析其表达与临床病理特征的关系。结果PCDH9在前列腺癌组织中的阳性率(72.8%)低于良性前列腺组织(100%)(P<0.05);前列腺癌组织中STAT3、Cyclin D1及C-myc的阳性率分别为82.6%、68.5%、64.1%。不同WHO/ISUP分级分组、PSA水平、临床分期的前列腺癌组间PCDH9的阳性率差异有显著性(P<0.05),高WHO/ISUP分级分组、PSA>20 ng/mL及临床分期Ⅲ+Ⅳ期组前列腺癌患者的PCDH9阳性率明显降低。STAT3表达与WHO/ISUP分级分组、PSA水平相关(P<0.05)。PCDH9表达与STAT3、Cyclin D1、C-myc表达均呈负相关(r值分别为-0.216、-0.414、-0.457,P均<0.05)。结论PCDH9表达缺失前列腺癌中存在STAT3过表达,进一步提示STAT3信号通路在介导前列腺癌中PCDH9表达缺失、抑制肿瘤细胞凋亡和促进肿瘤进展中具有重要作用。Purpose To analyze the role of PCDH9 in regulating the cycle of prostate cancer cells,and to explore the molecular mechanism of its deletion in inhibiting apoptosis and promoting progression of prostate cancer cells.Methods The expressions of PCDH9,STAT3,Cyclin D1 and C-myc in 92 cases of prostate cancer and 36 cases of benign prostate tissue were detected by immunohistochemical staining,and their correlation with clinicopathological characteristics was analyzed.Results The positive rate of PCDH9 in prostate cancer tissues(72.8%)was significantly lower than that in benign prostate tissues(100%)(P<0.05).The positive expression rates of STAT3,Cyclin D1 and C-myc in prostate cancer tissues were 82.6%,68.5% and 64.1%,respectively.The positive expression rate of PCDH9 in prostate cancer patients with different WHO/ISUP classification group,PSA level and clinical stage was different(P<0.05).The positive rate of PCDH9 in prostate cancer patients with high WHO/ISUP classification group,PSA>20 ng/mL and Ⅲ+Ⅳ group was significantly reduced.The expression of STAT3 was correlated with WHO/ISUP classification and PSA level(P<0.05).In 23 of 76 cases prostate cancer tissues with STAT3 positive expression,PCDH9 protein expression was lost.The expression of PCDH9 was negatively correlated with STAT3,Cyclin D1,and C-myc expression in prostate cancer tissues(r=-0.216,-0.414 and-0.457,P<0.05).Conclusion The presence of STAT3 overexpression in PCDH9 expression-dificient prostate cancer which further suggests that STAT3 signaling pathway plays an important role in mediating PCDH9 dificiency,inhibiting tumor cell apoptosis and promoting tumor progression in prostate cancer.
