摘  要:目的探究电针足三里对术后肠麻痹小鼠肠动力减低的改善作用及机制。方法将30只C57BL/6雄性小鼠随机分为假手术组、模型组和电针组。模型组和电针组采用挤压法进行术后肠麻痹造模,电针组于术后24 h开始连续电针足三里5 d。干预结束后,检测3组小鼠肠动力(结肠排珠时间、肠运输时间、粪便干重及干湿比);取小鼠结肠组织,透射电镜观察肠神经元线粒体形态、数量和体积,采用激光共聚焦显微镜观察线粒体膜电位变化,采用萤火虫荧光素酶催化荧光素发光法检测线粒体ATP含量,采用Western blot法检测线粒体生物发生相关蛋白Nrf-1、TFAM、COX-Ⅳ表达情况。结果与假手术组比较,模型组小鼠结肠排珠时间、肠运输时间均明显延长(P均<0.05),粪便干重明显降低(P<0.05),粪便液体百分比明显增高(P<0.05);与模型组比较,电针组小鼠结肠排珠时间、肠运输时间明显缩短(P均<0.05),粪便干重增加(P<0.05),粪便液体百分比减少(P<0.05)。与假手术组比较,模型组小鼠线粒体数量明显减少(P<0.05),线粒体体积明显增大(P<0.05),线粒体膜电位、线粒体ATP含量及Nrf-1、TFAM、COX-Ⅳ蛋白相对表达量均明显降低(P均<0.05);与模型组比较,电针组小鼠线粒体形态明显改善,线粒体数量明显增加(P<0.05),线粒体体积明显缩小(P<0.05),线粒体膜电位、线粒体ATP含量及Nrf-1、TFAM、COX-Ⅳ蛋白相对表达量均明显增高(P均<0.05)。结论电针足三里可明显改善术后肠麻痹小鼠的肠动力,其可能通过上调肠神经元线粒体生物发生相关蛋白Nrf-1、TFAM、COX-Ⅳ的表达,从而增加线粒体数量,抑制线粒体肿胀,提高线粒体的膜电位和ATP含量,改善线粒体能量代谢障碍起作用。Objective It is to explore the improving effect and mechanism of electroacupuncture at Zusanli on decreased intestinal motility in mice with postoperative intestinal paralysis.Methods Thirty C57BL/6 male mice were randomly divided into sham operation group,model group and electroacupuncture group.The model group and the electro-acupuncture group were squeezed for establishing models of postoperative intestinal paralysis,and the electro-acupuncture group was continuously electro-acupunctured at Zusanli for 5 days at 24 hours after the operation.After the intervention,the intestinal motility(colon bead discharge time,intestinal transit time,fecal dry weight and dry-to-wet ratio)of the mice in the three groups were detected;the mouse colon tissue was taken to observe the morphology,number and volume of enteric neuron mitochondria by transmission electron microscope,the changes of mitochondrial membrane potential were observed by laser confocal microscopy,the content of mitochondrial ATP was detected by firefly luciferase-catalyzed luciferin luminescence method,and the expression of mitochondrial biogenesis-related proteins Nrf-1,TFAM,and COX-Ⅳwere detected by Western blot method.Results Compared with the sham operation group,the colonic bead discharge time and intestinal transit time were significantly prolonged in the model group(all P<0.05),the dry weight of feces was significantly reduced(P<0.05),and the percentage of fecal fluid was significantly increased(P<0.05).Compared with the model group,the colonic bead discharge time and intestinal transit time were significantly shortened(all P<0.05),the dry weight of feces was significantly increased(P<0.05),and the percentage of fecal fluid was significantly decreased in the electroacupuncture group(P<0.05).Compared with the sham operation group,the number of mitochondria in the model group was significantly reduced(P<0.05),and the volume of mitochondria was significantly increased(P<0.05),the mitochondrial membrane potential,mitochondrial ATP content and the relati
