摘 要:目的通过在家兔腓肠肌急性钝挫伤模型中,研究Wnt1、Wnt4的含量变化,探讨Wnt1、Wnt4在急性钝挫伤修复中产生的作用。方法将40只日本大耳白兔随机选5只为空白对照组,其余35只通过软组织钝挫伤重力砸伤器建立家兔右后肢急性钝挫伤模型,造模成功后(4 h后)选取7个时间观察点进行取材,分别在第1天、2天、3天、4天、5天、7天、14天取材。通过透射电镜观察超微结构下各组急性钝挫伤后恢复情况。采用RT-PCR检测各组Wnt1 mRNA、Wnt4 mRNA表达动态变化情况。结果透射电镜下空白组、急性钝挫伤后各组可见不同程度的超微结构变化。RT-PCR检测结果显示,Wnt1 mRNA在损伤后7 d、14 d表达升高(P<0.05);Wnt4 mRNA在损伤后5 d、7 d、14 d表达升高(P<0.05)。结论Wnt1、Wnt4可能参与了兔急性钝挫伤后的早期恢复过程,且Wnt1、Wnt4的表达量变化与骨骼肌急性钝挫伤的早期恢复进程密切相关。说明Wnt1、Wnt4在骨骼肌损伤后的修复过程中发挥了重要作用。Objective To investigate the effect of Wnt1 and Wnt4 on the repair of acute gastrocnemius contusion in rabbits.Methods 40 Japanese big ear white rabbits were selected,5 rabbits were selected as blank control group by random number table method,and the other 35 rabbits were made acute contusion model of right hind limb by soft tissue contusion gravity smasher.After successful modeling(4 hours later),7 time observation groups were selected for sampling,which were 1d,2d,3d,4d,5d,7d and 14d respectively.The recovery of acute contusion was observed by transmission electron microscope.RT-PCR was used to detect the expression of Wnt1 mRNA and Wnt4 mRNA.Results The ultrastructural changes of blank group and acute contusion group were observed under transmission electron microscope.RT-PCR results showed that the expression of Wnt1 mRNA increased at 7 d and 14 d after injury(P<0.05);The expression of Wnt4 mRNA increased on the 5th,7th and 14th day after injury(P<0.05).Conclusion Wnt1 and Wnt4 may be involved in the early recovery process after acute contusion in rabbits,and the expression of Wnt1 and Wnt4 is closely related to the early recovery of acute contusion in skeletal muscle.These results indicate that Wnt1 and Wnt4 play an important role in the repair of skeletal muscle injury.
