摘 要:目的探讨甲氟喹对急性癫痫小鼠认知功能的影响。方法将30只C57 Thy1-YFP小鼠随机分为癫痫组、甲氟喹组和对照组。前2组小鼠采用匹鲁卡品腹腔注射建立癫痫模型,建模前甲氟喹组腹腔注射甲氟喹注射液,其余2组腹腔注射0.9%氯化钠注射液。建模成功后利用Morris水迷宫实验测定并比较3组小鼠的认知功能,主要指标包括逃避潜伏期和穿越平台次数。结果定位航行试验第1天,癫痫组、甲氟喹组小鼠逃避潜伏期均明显长于对照组(均P<0.05),而癫痫组与甲氟喹组比较差异无统计学意义(P>0.05);第2~5天,癫痫组、甲氟喹组小鼠逃避潜伏期均明显长于对照组(均P<0.05),但甲氟喹组均短于癫痫组(均P<0.05)。第6天空间探索试验中,癫痫组、甲氟喹组小鼠穿越平台次数均明显少于对照组(均P<0.05),但甲氟喹组多于癫痫组(P<0.05)。结论甲氟喹控制小鼠癫痫发作的同时,还能改善其空间学习及记忆能力,显著提高癫痫小鼠的认知功能。Objective To investigate the effects of mefloquine on cognitive function in acute epileptic mice.Methods Thirty C57 Thy1-YFP mice were randomly divided into epilepsy group,mefloquine group and control group.Epilepsy mouse model were established by intraperitoneal injection of pilocarpine.The mefloquine group was intraperitoneal injected with mefloquine before modeling,while the other 2 groups were injected with 0.9%sodium chloride.The cognitive functions of mice,including escape latency and the number of crossing platform were measured by Morris water maze test.Results On d1 of the navigation test,the escape latency of the epileptic and mefloquine group was significantly longer than control group(both P<0.05),and there was no significant difference between epileptic and mefloquine groups(P>0.05).On d2-d5,compared with the control group,the escape latency was significantly longer in epileptic and mefloquine group(all P<0.05),but it was shorter in mefloquine group than that in epilepsy group(all P<0.05).On d6,the times of crossing platform in space exploration test of epileptic and mefloquine groups were significantly less than those of control group(both P<0.05),while the times of crossing platform in mefloquine group were more than those in epilepsy group(P<0.05).Conclusion Mefloquine can improve the ability of spatial learning and memory as well as the cognitive function of epileptic mice.
