摘 要:试验以超声波辅助提取法提取沧州海兴湿地二色补血草(Limonium bicolor)中的总黄酮。在单因素试验基础上,采用Box-Behnken中心组合设计响应面试验,优化二色补血草总黄酮的提取工艺,并进行稳定性和抑菌性研究。结果显示,响应面法优化二色补血草总黄酮的最佳提取条件为超声功率250 W、料液比1∶35、乙醇浓度51%。此条件下,二色补血草总黄酮提取率为3.32%,与预测值3.41%接近。二色补血草总黄酮在低温及避光条件下稳定性表现较好,且对大肠杆菌、枯草芽孢杆菌、黑曲霉和毛霉具有不同程度的抑菌作用,最小抑菌浓度为0.162 g/L。随着浓度增加,二色补血草总黄酮对枯草芽孢杆菌和毛霉的抑菌效果更为明显。研究表明,二色补血草总黄酮稳定性较好,可作为新型天然抑菌剂应用于饲料添加剂领域。The experiment extracted total flavonoids from Limonium bicolor in Haixing wetland of Cangzhou by ultrasonic-assisted extraction method. On the basis of single factor experiment, the experiment used the Box-Behnken central combination to design response surface experiment to optimize the total flavonoids extraction technology, and the stability and antibacterial activity were studied. The results showed that the best extraction conditions of total flavonoids of Limonium bicolor optimized by response surface were ultrasonic power 250 W, material to liquid ratio 1∶35 and ethanol concentration 51%. Under the above conditions, the extraction rate of total flavonoids was 3.32%, which was near with the predicted value of 3.41%. The total flavonoids of Limonium bicolor showed good stability under low temperature and dark conditions, and had different inhibitory effect on Escherichia coli, Bacillus subtilis, Aspergillus niger and Mucor. The minimum inhibitory concentration was 0.162 g/L, and the inhibitory effect on Bacillus subtilis and Mucor was more obvious with the increase of concentration. The experiment indicates that the total flavonoids of Limonium bicolor has good stability, and can be used as a new natural antibacterial agent in the field of feed additives.
