摘 要:目的分析褪黑素与人胃癌细胞生长指标的关系,探讨褪黑素对人胃癌细胞增殖、肿瘤生长的影响。方法选用人胃癌细胞株MGC-803、SGC-7901及AGS培养、传代,在氯化钴(CoCl_(2))模拟乏氧条件下及常氧条件下,采用0.01、0.1、1、3 mmol/L不同浓度的褪黑素干预,分别在不同时间点0.5、2、16、24 h给药,观察褪黑素对人胃癌细胞生物学行为的影响。结果常氧及乏氧条件下,低浓度褪黑素(0.01、0.1 mmol/L)对胃癌细胞无明显抑制作用;而高浓度褪黑素(1、3 mmol/L)能明显抑制人胃癌细胞株MGC-803、SGC7901及AGS增殖,呈时间和剂量依赖趋势。各组间癌细胞抑制率比较,差别有统计学意义(P<0.05)。3 mmol/L褪黑素在体外抑制人胃癌细胞增殖最为显著,并能明显抑制SGC-7901细胞分泌血管内皮生长因子-A(VEGF-A)。结论常氧及乏氧条件下,褪黑素体外能抑制人胃癌细胞株MGC-803、SGC-7901及AGS增殖,呈时间-剂量依赖性,高浓度褪黑素明显抑制SGC-7901细胞分泌VEGF-A。Objective Analyze the relationship between melatonin and the growth indices of human gastric cancer cells,and explore the effect of melatonin on the proliferation and tumor growth of human gastric cancer cells.Methods Human gastric cancer cell lines MGC-803,SGC-7901 and AGS were cultured and subcultured.Under hypoxic and normoxic conditions simulated by cobalt chloride(CoCl_(2)),different concentrations of melatonin were administered with 0.01,0.1,1 and 3 mmol/L,and blank control group was set in 0.5,2,16 and 24 h at different time points,the effect of melatonin on biological behavior of human gastric cancer cells were observed.Results Low concentrations of melatonin(0.01 and 0.1 mmol/L)in normoxic and hypoxic conditions had no significant inhibitory effect on gastric cancer cells,while high concentrations of melatonin(1 and 3 mmol/L)significantly inhibited the proliferation of human gastric cancer cells MGC-803,SGC-7901 and AGS in a time-dependent and dose-dependent trend.There was statistically significant difference in the inhibition rate of cancer cells among all groups(P<0.05).Melatonin of 3 mmol/L inhibited the proliferation of human gastric cancer cells significant and VEGF-A secretion of SGC-7901 cells in vitro.Conclusion In vitro melatonin can inhibit the proliferation of human gastric cancer cell lines MGC-803,SGC-7901 and AGS in a time-dose dependent manner under normoxic and hypoxic conditions.High concentration of melatonin can significantly inhibit VEGF-A secretion in SGC-7901 cells.
