摘 要:目的阐明网状纤维染色中银氨离子[Ag(NH_(3))_(2)^(+)]在蒸馏水中的解离规律,提高网状纤维染色的稳定性及一致性。方法收集江门市中心医院病理科10例肝脏手术切除石蜡标本,采用改良Gordon-Sweets法进行染色,银氨液反应后置于蒸馏水中浸洗不同的时间以获得最佳的解离点,并在中山大学肿瘤防治中心146例肝癌病例中进行验证。结果网状纤维染色中银氨液完全反应后在蒸馏水中浸洗2min时网状纤维染色强,定位准确,且背景干净,信噪比最佳,优片率达98.6%(144/146);2min后结合Ag(NH_(3))_(2)^(+)开始逐步解离,15min后基本完全解离,镜下网状纤维变成紫红色,不能满足病理诊断。结论网状纤维染色中结合的Ag(NH_(3))_(2)^(+)在蒸馏水中浸洗2min后呈逐步解离,建议蒸馏水浸洗时间控制在1~2min,可获最佳效果,满足病理诊断。Objective To clarify the dissociation pattern of Ag(NH_(3))_(2)^(+) in distilled water during reticular fiber staining,and improve the stability and consistency of the staining.Methods Paraffin samples of 10 cases of liver resection were collected from the pathology department of Jiangmen Central Hospital,and the modified Gordon-Sweets method was used for staining.After reaction in the silver ammonia solution,the samples were soaked in distilled water for different periods to obtain the optimal dissociation point.The obtained pattern was then verified on 146 cases of liver cancer samples in Sun Yat-sen University Cancer Center.Results In the reticular fiber dyeing,when the silver ammonia solution is completely reacted,the samples immersed in distilled water for 2 minutes showed the strongest staining and the background was clean.The excellent rate reached 98.6%(144/146),which could meet the standard of pathological diagnosis.After 2 minutes,the Ag(NH_(3))_(2)^(+) complex began to dissociate,and then completely dissociated in 15minutes with the stained fibers becoming dark purple and not suitable for pathological diagnosis.Conclusion The Ag(NH_(3))_(2)^(+) dissociates slowly after 2 minutes in distilled water.The best staining result suitable for pathological diagnosis can be obtained by soaking samples in distilled water for 1 to 2 minutes.
