摘  要:斑马鱼是一种脊椎模式生物,已成为生命科学研究的新宠,被广泛用于构建人类的各种疾病模型,建立药物筛选和治疗的研究平台。研究以5 dpf斑马鱼为对象对不同浓度葡萄糖、四氧嘧啶及其组合对斑马鱼葡萄糖水平的影响进行系统研究,探讨建立了基于生理和病理生理的糖代谢异常斑马鱼生物模型,在此基础上对桑叶和绞股蓝叶水提取物的降糖作用进行比较研究。以高浓度葡萄糖(22、26、30 mg/mL)溶液、四氧嘧啶(0.04、0.06、0.08、0.10 mM)药物、葡萄糖(22 mg/mL)溶液与四氧嘧啶(0.02、0.04、0.08 mM)药物联合可显著增加斑马鱼葡萄糖水平,成功建立基于生理和病理生理的糖代谢异常斑马鱼模型。同时研究发现,200、400和800μg/mL桑叶和800μg/mL绞股蓝叶水提取物可以显著降低高糖诱导型斑马鱼的葡萄糖水平,200μg/mL桑叶和400μg/mL绞股蓝叶水提取物可以显著降低胰岛损伤型斑马鱼的葡萄糖水平。基于斑马鱼生物模型评价,桑叶和绞股蓝水提取物均具有降糖作用,但在同等剂量下桑叶的降糖效应明显强于绞股蓝,两者在辅助降血糖功能食品开发上具有广阔的应用前景。Zebrafish is a biological model of vertebrate,which has become a new favorite in life science.It is widely used to establish models of various human diseases and to set up a research platform for drug screening and treatment.In this study,5 dpf zebrafish were selected to systematically study the effects of different concentrations of glucose,alloxan and their combinationon the level of glucose of zebrafish,and zebrafish as a biological model with glucose metabolism disorders based on physiology and pathophysiology was established.On this basis,the hypoglycemic effects of the aqueous extract of moras folium and gynostemma pentaphyllum leaves were evaluated.The results showed that high concentration of glucose(22,26,30 mg/mL),alloxan(0.04,0.06,0.08,0.10 mM),and the combination of glucose(22 mg/mL)and alloxan(0.02,0.04,0.08 mM)significantly increased the level of glucose of zebrafish.Therefore,a model of glucose metabolism disorders in zebrafish was established.Meanhilw,the research showed that 200,400 and 800μg/mL of the aqueous extract of moras folium and 800μg/mL of the aqueous extract of gynostemma pentaphyllum leaves significantly reduced the level of glucose of high glucose-induced zebrafish.And 200μg/mL of the aqueous extract of mulberry leaves and 400μg/mL of the aqueous extract of gynostemma pentaphyllum leaves significantly reduced the level of glucose of zebrafish with damaged pancreatic islets.The results also showed that the aqueous extract of moras folium and gynostemma pentaphyllum leaves had hypoglycemic effects,but the hypoglycemic effect of the aqueous extract of mulberry leaves was significantly stronger than the aqueous extract of gynostemma pentaphyllum leaves at the same concentration,making them have broad prospect of application in the development of functional food in assistant decreasing blood glucose.
