摘 要:目的探讨317L-Cu抗菌不锈钢(317L-Cu SS)对小鼠成纤维细胞(L-929细胞)增殖、黏附、凋亡等生物学行为的影响,评价抗菌不锈钢材料的细胞毒性;同时观察不同浓度Cu^(2+)对细胞毒性的影响。方法检测L-929细胞在普通医用不锈钢(317L SS)、317L-Cu SS、医用纯钛(Ti)及纯铜(Cu)材料浸提液中培养24 h、48 h、72 h时的光密度(OD)值,判断细胞毒性级别;应用流式细胞仪Annexin V-FITC/PI双染法检测细胞凋亡情况;吖啶橙染色法检测L929细胞在材料表面的黏附情况。结果317L-Cu SS组24 h、72 h OD值、细胞黏附数量均低于Ti组而高于317L SS组(均P<0.05)。317L-Cu SS组细胞毒性分级为1级,Cu组细胞毒性分级为3级或4级;与317L SS组比较,317L-Cu SS组细胞凋亡率显著降低(P<0.05)。而与阴性对照组、Ti组比较,317L-Cu SS组细胞凋亡率无统计学差异(均P>0.05)。纯铜组细胞凋亡率明显高于其他各组(均P<0.05)。结论317L-Cu抗菌不锈钢对细胞的增殖、凋亡、黏附等生物学行为无明显不良影响,生物相容性良好,而纯铜材料细胞毒性明显,Cu^(2+)的细胞毒性呈双向性。Objective To observe the effects of 317L-Cu antibacterial stainless steel(317L-Cu SS)on the biological behaviors of mouse fibroblasts(L-929 cells),including proliferation,adhesion,and apoptosis;evaluate the cytotoxicity of antibacterial stainless steel;and observe the effects of different concentrations of Cu^(2+)on cytotoxicity.Methods The optical density(OD)values of the L-929 cells were measured at 24 h,48 h,and 72 h in ordinary medical-grade stainless steel(317L SS),317L-Cu SS,medical pure titanium(Ti),and pure copper(Cu)material extracts to determine the cytotoxicity level.The flow cytometry Annexin V-FITC/PI double staining method was applied to detect apoptosis.Acridine orange staining was applied to detect the adhesion of L929 cells on the material.Results The 24 h and 72 h OD values and cell adhesion numbers were lower in the 317L-Cu SS group than in the Ti group(all P<0.05)and higher than those of the 317L SS group(P<0.05).The 317L-Cu SS group had a cytotoxicity grade of 1,while the Cu group had cytotoxicity grades of 3 or 4.Apoptosis rate was significantly lower in the 317L-Cu SS group than in the 317L SS group(P<0.05).There was no significant difference in the apoptosis rate of the 317L-Cu SS group versus the negative control and Ti groups(all P>0.05).The apoptosis rate was significantly higher in the Cu group than in the other groups(P<0.05).Conclusion The 317L-Cu SS material was biocompatible and had no significant adverse effects on biological behaviors such as cell proliferation,apoptosis,and adhesion,whereas the cytotoxicity of the Cu material was significant and that of Cu^(2+)was bidirectional.
