调衡方对Lewis肺癌荷瘤小鼠瘤组织与肺组织Foxp3、IL-10、TGF-β表达的影响

摘　　要：目的研究调衡方对Lewis肺癌荷瘤小鼠血清白细胞介素-10(IL-10)、转化生长因子-β(TGF-β)水平及瘤组织、肺脏组织中Foxp3、IL-10、TGF-β表达的影响,探讨调衡方对荷瘤体免疫抑制细胞的调节机制。方法将60只雄性C57BL/6小鼠随机分为正常组、荷瘤组、贞芪组及调衡方高、中、低剂量组,每组10只。除正常组外,其余组均将传代培养的Lewis肺癌细胞接种于小鼠右腋皮下建立肺癌模型。接种24 h后,贞芪组给予贞芪扶正胶囊41.5 g/kg灌胃,调衡方低、中、高剂量组分别给予调衡方流浸膏12.5 g/kg、25 g/kg、50 g/kg灌胃,荷瘤组给予等容量生理盐水灌胃,均1次/d,连续8 d。灌胃结束后,HE染色观察瘤组织及肺组织细胞形态,ELISA法检测血清IL-10、TGF-β水平,免疫组化法检测瘤组织及肺组织中Foxp3、IL-10、TGF-β表达情况。结果HE染色显示:荷瘤组小鼠瘤组织中瘤细胞坏死面积很小,有少量的凋亡瘤细胞;肺组织可见细胞排列混乱的肿瘤转移灶,病理性的核分裂较多,肿瘤实、间质间边缘不清。调衡方各组瘤组织中多处瘤细胞核固缩,瘤细胞胞膜皱缩不完整,瘤细胞残留坏死物随处可见,尤其是调衡方高、中剂量组明显;调衡方低剂量组肺组织可见肿瘤细胞,调衡方中、高剂量组未见明显的肿瘤细胞聚集,且调衡方高剂量组细胞排列与正常肺组织形态接近。荷瘤组血清IL-10、TGF-β水平均明显高于正常组(P均<0.05);贞芪组和调衡方各组血清IL-10、TGF-β水平均明显低于荷瘤组(P均<0.05),调衡方高剂量组均明显低于调衡方低、中剂量组(P均<0.05);调衡方各组血清IL-10水平与贞芪组比较差异均无统计学意义(P均>0.05),调衡方高剂量组血清TGF-β水平明显低于贞芪组(P<0.05)。贞芪组和调衡方各组瘤组织及肺组织中Foxp3、IL-10、TGF-β蛋白表达阳性率均明显低于荷瘤组(P均<0.05),调衡方高剂量组均明显低于调衡方低剂量组(P�Objective It is to study the effects of Tiaoheng Decoction on the levels of serum interleukin-10(IL-10)and transforming growth factor-β(TGF-β)and the expression of Foxp3,IL-10 and TGF-βin tumor tissues and lung tissues of Lewis lung cancer-bearing mice,and to explore the regulatory mechanism of Tiaoheng Decoction on immunosuppressive cells of tumor-bearing body.Methods Sixty male C57BL/6 mice were randomly divided into normal group,tumor-bearing group,Zhenqi group and high,medium and low dose Tiaoheng Decoction groups,with 10 mice in each group.Except for the normal group,the other groups were inoculated with subcultured Lewis lung cancer cells into the right axillary skin of mice to establish lung cancer models.At 24 hours after inoculation,Zhenqi group was given Zhenqi Fuzheng capsule 41.5 g/kg by gavage,and low,medium and high dose Tiaoheng Decoction groups were respectively given Tiaoheng Decoction liquid extract 12.5 g/kg,25 g/kg,50 g/kg by gavage,the tumor-bearing group were given the same volume of normal saline by gavage,all once a day,continuously treated for 8 days.After the treatment,the cell morphology of tumor tissue and lung tissue were observed by HE staining,the levels of serum IL-10 and TGF-βwere detected by ELISA,the expression of Foxp3,IL-10,TGF-βin tumor tissue and lung tissue was detected by immunohistochemical method.Results HE staining showed that the necrosis area of tumor cells in tumor tissue of tumor-bearing group was very small,and there were a few apoptotic tumor cells;tumor metastases with disordered cell arrangement were seen in lung tissue,and there were more pathological nuclear divisions,and the edges between tumor parenchyma and stroma were not clear.There were many tumor cell karyopyknosis with incomplete cell membrane shrinkage and residual necrosis of tumor cells everywhere in the tumor tissues of each group of Tiaoheng Decoction,especially in the high and medium dose groups of Tiaoheng Decoction;tumor cells could be found in the lung tissue of the low-dose group of Tiao
