摘 要:低温寒害是制约我国橡胶树种植业发展的关键因素,为了增加国内橡胶自给率,培育抗寒能力强的无性系以扩大种植面积是有效途径之一。本研究以云南培育的三倍体无性系“云研77-4”为材料,采用PacBio测序技术对不同时间低温胁迫处理的混合样进行全长转录组测序及分析,共产出41.85 Gb的clean data数据,分析得到高质量转录本154484条,平均每条序列长度为2181 bp,其中新转录本70055条。经对新发现转录本的SSR、编码区序列预测、lncRNA及转录因子分析,并对新转录本进行功能注释,共发现80432个SSRs,66182个ORF,66684个功能基因,预测到转录因子12064个,其中包含了与抗逆性相关的MYB、C3H转录因子。本研究可为进一步筛选抗寒性相关的转录因子和抗寒三倍体育种提供参考。Hevea brasiliensis,as a main source of natural rubber,is the most important tropical crop in China.Low temperature is a key factor restricting the development of rubber tree planting industry in China.In order to increase the domestic rubber self-sufficiency rate,it is one of the effective ways to cultivate clones with strong cold resistance clones to expand the planting area.In this study,the full-length transcriptome sequence of mixed samples of Yunyan 77-4 with different low temperature stress was sequenced by PacBio transcriptome sequencing technology and the sequences were analyzed.The results showed that 41.85Gb clean data and 154484 high quality transcript were obtained,and the average length of each sequence was 2181bp,the new transcript was 70055.The SSR,coding regions,lncRNA and transcription factors and functionally comments were analyzed,a total of 80432 SSRs,66182 ORFs,66684 functional genes and 12064 transcription factors containing MYB,C3H transcription factors associated with inversion resistance were obtained.The results will provide a reference for further screening of cold-related transcription factors and cold-resistance triploid breeding.
