摘　　要：目的基于磷酸肌醇-3激酶(PI3K)/蛋白激酶B(Akt)信号通路研究幽门螺杆菌肿瘤坏死因子-α诱导蛋白(Tipα)对胃癌细胞顺铂敏感性的作用机制。方法使用Tipα刺激胃癌SGC-7901细胞,采用CCK8检测细胞活性,流式细胞术检测细胞凋亡,Western blot检测PI3K/Akt信号通路相关蛋白——PI3K、p-PI3K、Akt、p-Akt表达水平,运用PI3K/Akt抑制剂——LY294002预处理细胞阻断PI3K/Akt信号通路,探讨PI3K/Akt信号通路活性与Tipα对胃癌细胞顺铂敏感性作用的关系。结果与顺铂组比较,Tipα联合顺铂组细胞活性、侵袭力、p-PI3K/PI3K比值、p-Ak/Akt比值均明显增加,细胞凋亡率明显降低,差异均有统计学意义(P<0.05);LY294002预处理可逆转Tipα对胃癌细胞顺铂敏感性的抑制作用。结论Tipα可通过激活PI3K/Akt信号通路抑制胃癌细胞的顺铂敏感性。Objective To study the role mechanism of Helicobacter pylori Tipαprotein on the sensitivity of gastric cancer cells to cisplatin based on the PI3K/Akt signaling pathway.Methods The gastric cancer SGC-7901 cells were stimulated by using Tipα,the cell viability was detected by using CCK8;the cell apoptosis was detected by using flow cytometry;the expression levels of PI3K/Akt signaling pathway related proteins PI3K,p-PI3K,Akt,and p-Akt were detected by using the Western blot;the PI3K/Akt inhibitor LY294002 was used to pretreat the cells to block the PI3K/Akt signaling pathway,and then the relationship between the activity of PI3K/Akt signaling pathway and the effect of Tip protein on the sensitivity of gastric cancer cells to cisplatin was investigated.Results Compared with the cisplatin group,the cell viability,invasion ability,p-PI3K/PI3K ratio value and p-Ak/Akt ration value of the Tipαcombined with cisplatin group were significantly increased,the apoptosis rate was significantly reduced,and the differences were statistically significant(P<0.05);the LY294002 pretreatment could reverse the inhibitory effect of Tipαon the sensitivity of gastric cancer cells to cisplatin.Conclusion Tipαcould inhibit the cisplatin sensitivity of gastric cancer cells by activating the PI3K/Akt signaling pathway.
