摘  要:目的探讨肿瘤抑素19肽对肝癌SK-Hep-1细胞增殖能力和凋亡作用的影响。方法CCK-8检测不同浓度19肽(0、50、100、150、200、250μg/ml)对SK-Hep-1细胞增殖能力的影响。DAPI染色检测不同浓度19肽作用SK-Hep-1细胞凋亡形态学变化,流式细胞术检测不同浓度的19肽对SK-Hep-1细胞凋亡的影响。结果0、50、100、150、200、250μg/ml浓度的19肽作用SK-Hep-1细胞24 h后细胞存活率分别为(100.00±1.80)%、(95.64±1.92)%、(82.18±1.67)%、(68.43±1.39)%、(46.74±1.35)%、(42.49±1.85)%。作用48 h后细胞存活率分别为(100.00±1.80)%、(87.27±1.96)%、(66.32±1.74)%、(58.78±1.94)%、(40.40±1.44)%、(22.15±1.51)%。19肽为0μg/ml时,SK-Hep-1细胞大部分染色均匀,呈蓝色,细胞未发生凋亡。与0μg/ml相比,50、100、150、200、250μg/ml浓度的19肽作用24 h后,部分细胞呈亮蓝色,细胞核出现浓缩、破裂、碎块状致密浓染或帽点状凋亡形态。细胞凋亡率随着19肽浓度的增高而增加。结论19肽能有效抑制SK-Hep-1细胞的增殖,诱导SK-Hep-1细胞的凋亡,为新型抗肝癌药的研究和临床治疗提供理论指导。Objective To study the effect of tumstatin 19 peptide proliferation and apoptosis of hepatoma SK-Hep-1 cells.Methods CCK-8 was used to detect the effect of 19 peptide(0,50,100,150,200,250μg/ml)on the proliferation of SK-Hep-1 cells.DAPI staining was used to detect the morphological changes of SK-Hep-1 cell apoptosis,and flow cytometry was used to detect the effect of 19 peptide on SK-Hep-1 cell apoptosis.Results Sk-hep-1 cells were treated with 0,50,100,150,200,250μg/ml 19 peptide,after 24 h,the survival rates were(100.00±1.80)%,(95.64±1.92)%,(82.18±1.67)%,(68.43±1.39)%,(46.74±1.35)%,(42.49±1.85)%,respectively.After 48 h treatment,the cell survival rate was(100.00±1.80)%,(87.27±1.96)%,(66.32±1.74)%,(58.78±1.94)%,(40.40±1.44)%,(22.15±1.51)%,respectively.When 19 peptide was 0μg/ml,most of SK-HEP-1 cells were stained uniformly and blue,and apoptosis did not occur.Compared with 0μg/ml,some cells showed bright blue after treated with 50,100,150,200 and 250μg/ml 19 peptide for 24 h,and the nuclei showed concentration,rupture,fragmentary dense and dense staining or cap-like apoptotic morphology.The apoptosis rate increased with the increase of 19 peptide concentration.Conclusion 19 peptide can effectively inhibit the proliferation of SK-Hep-1 cells and induce the apoptosis,which provides theoretical guidance for the research and clinical treatment of new anti hepatoma drugs.
