摘 要:目的:探讨Rpph1对高糖作用的肾小管上皮细胞生物行为的影响。方法:将HK2细胞随机分为对照组、高糖组、高糖+pcDNA3.1组、高糖+pcDNA3.1-Rpph1组、高糖+si-NC组、高糖+si-Rpph1组、高糖+si-Rpph1+si-NC组、高糖+si-Rpph1+si-HuR组。实时荧光定量PCR(RT-qPCR)检测Rpph1和HuR mRNA表达水平;蛋白质印迹(Western blot)法检测蛋白表达;四甲基偶氮唑盐比色法(MTT)检测细胞存活率;流式细胞术检测细胞凋亡。结果:高糖作用的肾小管上皮细胞中Rpph1、HuR表达水平升高,P21、Caspase-3、Vimentin表达水平升高,E-cadherin表达水平降低,细胞存活率降低,细胞凋亡率升高(P<0.05)。过表达Rpph1促进细胞凋亡,抑制细胞存活;而干扰Rpph1表达与过表达Rpph1的作用相反。抑制HuR表达增强了抑制Rpph1对高糖作用的肾小管上皮细胞存活率、凋亡和上皮间质转化(EMT)的影响。结论:干扰Rpph1表达可能通过下调HuR抑制高糖作用的肾小管上皮细胞凋亡和EMT。Objective:To investigate the effect of Rpph1 on the biological behavior of renal tubular epithelial cells with high glucose.Methods:The HK2 cells were randomly divided into control group,high glucose group,high glucose+pcDNA3.1 group,high glucose+pcDNA3.1-Rpph1 group,high glucose+si-NC group,high glucose+si-Rpph1 group,high glucose+si-Rpph1+si-NC group,high glucose+si-Rpph1+si-HuR group.Real-time fluorescent quantitative PCR(RT-qPCR)was used to detect Rpph1 and HuR mRNA expressions;Western blot was used to detect protein expression;Tetramethylazozolium colorimetric method(MTT)to detect cell survival rate;flow cytometry to detect cell apoptosis.Results:In the renal tubular epithelial cells treated with high glucose,the expression levels of Rpph1 and HuR were increased,the expression levels of P21,Caspase-3,Vimentin were increased,the expression of E-cadherin was decreased,the cell survival rate was decreased,and the apoptosis rate was increased(P<0.05).Overexpression of Rpph1 promoted cell apoptosis and inhibited cell survival;and interference with Rpph1 expression has the opposite effect to overexpression of Rpph1.Inhibition of HuR expression enhanced the effect of inhibition of Rpph1 on the survival,apoptosis,and epithelial-mesenchymal transition(EMT)of renal tubular epithelial cells with high glucose action.Conclusion:Interfering with the expression of Rpph1 can inhibit apoptosis and EMT of high glucose renal induced tubular epithelial cells by down-regulating HuR.
