摘 要:目的研究高甘油三酯血症(hypertriglyceridemia,HTG)对大鼠急性胰腺炎(acute pancreatitis,AP)模型胰腺组织细胞间紧密连接(tight junctions,TJs)的影响。方法48只4周龄雄性SD大鼠随机分成普通饲养组(24只)和高脂饲养组(24只),4周后普通饲养组随机分为C组和AP组,高脂饲养组随机分为HTG组和高甘油三酯血症急性胰腺炎(HTGP)组。AP组和HTGP组经腹腔注射雨蛙肽建立AP模型,C组和HTG组经腹腔注射同等剂量的生理盐水,分别于造模24 h、48 h处置大鼠,采用HE染色法观察胰腺病理改变,免疫组织化学法检测胰腺组织中TJs蛋白脂解刺激脂蛋白受体(LSR)、Tricellulin蛋白(TRIC)、ZO-1、occludin、claudin-7定位及表达,透射电镜观察胰腺导管上皮细胞间TJs变化情况。结果高脂饲养组的大鼠血清甘油三酯(triglyceride,TG)较普通饲养组明显升高(P<0.05);HTG组胰腺组织病理评分高于C组(P<0.05),相同时点HTGP组胰腺组织病理评分高于AP组,但差异无统计学意义(P>0.05);HTG组LSR、TRIC表达显著低于C组(P<0.05),AP组和HTGP组的24 h时点LSR、TRIC、occludin、claudin-7表达均低于相应对照组(P<0.05),HTGP组24 h时点LSR和TRIC的表达显著低于AP组的对应时点(P<0.05);透射电镜观察AP组和HTGP组的24 h时点,发现主胰管上皮细胞TJs较C组和HTG组减少、细胞间隙增宽,且HTGP组比AP组的细胞间隙增宽更明显。结论TJs蛋白LSR、TRIC、ZO-1、occludin、claudin-7在HTG及HTGP大鼠胰腺组织中表达较非高脂模型下降,其中以三细胞间紧密连接(tricellular tight junctions,tTJs)蛋白LSR及TRIC下降明显,提示HTG可能减弱胰腺组织的tTJs,进而加重胰腺组织损伤。Objective To explore the effect of hypertriglyceridemia(HTG)in tight junctions(TJs)of pancreatic cell in acute pancreatitis(AP)rat.Methods Forty-eight four-week-old male Sprague-Dawley rats was randomly divided into normal chew fed group and high fatty chew fed group.Serum triglyceride(TG)was detected after 4 weeks of feeding.Then normal chew fed rats were divided into C group and AP group while high fatty chow fed rats were divided into HTG group and HTGP groups.Rats in AP group and HTGP group were intraperitoneally injected with caerulin to establish AP models,while rats in C group and HTG group were injected with the same volume of saline instead.Rats were sacrificed at 24 hours and 48 hours after injection.Pancreatic pathological changes were observed using hematoxylin-eosin(HE)staining.Expression levels of lipolysis-stimulated lipoprotein receptor(LSR),tricellulin protein(TRIC),ZO-1,occluding,and claudin-7 in the pancreas tissues were tested via immunohistochemistry.TJs changes of pancreatic ductal epithelial cells were observed using transmission electron microscope.Results Serum TG levels of rats in high fatty chew fed group were significantly higher than those of rats in normal chew fed group(P<0.05).Pancreatic pathological scores of HTG group were significantly higher than those of C group(P<0.05).Scores of HTGP group were higher than those of corresponding AP group though with no significant differences(P>0.05).In immunohistochemical analysis of pancreatic tissues,expressions of LSR and TRIC in HTG group were significantly decreased compared with C group(P<0.05).Decreased expressions of LSR,TRIC,occludin,and claudin-7 in both AP group and HTGP group at 24 hours were observed compared to C group and HTG group(P<0.05).Expressions of LSR and TRIC in HTGP group at 24 hours were significantly lower than those in AP group at 24 hours(P<0.05).Reduction or disappear of TJs and extended distance between cells were observed in modeling groups,especially in HTGP group at 24 hours,compared to C group and HTG group.
