摘  要:目的探讨miR-210-3p对宫颈癌细胞增殖、侵袭的影响及作用机制。方法通过OncomiR和GEPIA网站分析TCGA数据库宫颈癌病例中miR-210-3p和SMAD4的表达。采用miR-210-3p抑制物转染宫颈癌Hela细胞,实验设置空白组、miR-210-3p阴性对照组和miR-210-3p抑制物组,采用CCK-8法和Transwell实验分别检测Hela细胞的增殖和侵袭能力,采样Western blot检测Hela细胞SMAD4蛋白的表达,应用Targetscan7.2网站预测SMAD4是否为miR-210-3p的靶基因,应用双荧光素酶报告基因实验进行验证。结果 OncomiR和GEPIA网站分析结果显示,与正常组相比,宫颈癌组织中miR-210-3p表达增加,SMAD4的mRNA表达下降(P<0.05)。与miR-210-3p阴性对照组相比,miR-210-3p抑制物组细胞miR-210-3p表达下降(P<0.05),SMAD4蛋白表达升高(P<0.05),细胞增殖能力下降(P<0.05)、侵袭能力下降(P<0.05)。双荧光素酶实验显示SMAD4为miR-210-3p的靶基因。结论抑制miR-210-3p的表达可以抑制Hela细胞的增殖与侵袭,其机制可能与负调控SMAD4的表达相关。Objective To investigate the effect and mechanism of miR-210-3 p on the proliferation and invasion of cervical carcinoma cells. Methods OncomiR and GEPIA were used to analyze the expression of miR-210-3 p and SMAD4 in cervical carcinoma of TCGA database. The Hela cells were transfected, which were divided into blank control group(not transfected), miR-210-3 p negative control group(transfected unrelated sequence), and miR-210-3 p inhibitor group(transfected anti-miR-210-3 p). CCK-8 and Transwell were used to detect the proliferation and invasion, Western blot was used to detect the expression of SMAD4 protein, and whether SMAD4 was the target gene of miR-210-3 p was predicted by Targetscan 7.2, which was verified by double luciferase reporter gene experiment. Results OncomiR and GEPIA showed that the expression of miR-210-3 p was up-regulated and SMAD4 was down-regulated in cervical carcinoma compared with normal group. After miR-210-3 p was inhibited, the expression of SMAD4 was increased, and the proliferation and invasion ability of Hela cells were decreased. Bioinformatics analysis and dual luciferase reporter gene showed that SMAD4 was one of the target genes of miR-210-3 p. Conclusion miR-210-3 p may regulate the proliferation and invasion of Hela cells by targeting SMAD4.
