摘 要:目的探讨镁合金(Mg-Ag、Mg-Cu)对骨肉瘤细胞U2OS增殖、侵袭及迁移能力影响的机制。方法腐蚀实验评价Mg、Mg-Ag与Mg-Cu合金的降解速率;不同材料(Mg、Mg-Ag与Mg-Cu)浸提液处理组细胞的MTT及平板克隆形成实验检测骨肉瘤细胞增殖能力;Transwell侵袭与迁移实验评估细胞侵袭增殖能力;Western blot实验检测F-actin、β-catenin、Vimentin和N-cadherin的表达。结果 Mg-Ag与Mg-Cu合金的降解速率高于纯Mg;Mg-Ag与Mg-Cu合金组细胞增殖能力明显受到抑制;Mg-Ag与Mg-Cu合金组细胞的侵袭与迁移能力下降,并且F-actin、β-catenin、Vimentin和N-cadherin表达下调。结论 Mg-Ag与Mg-Cu合金可以通过Factin/β-catenin细胞信号通路调节细胞EMT,进而抑制骨肉瘤细胞U2OS的增殖、侵袭与迁移。Objective To investigate the effect of magnesium alloy(Mg-Cu and Mg-Ag) on proliferation,invasion and migration of osteosarcoma cell line U2 OS. Methods The degradation rate of Mg, Mg-Ag and Mg-Cu alloy was evaluated by corrosion assay. The proliferation ability of osteosarcoma cells was detected by MTT and plate clone formation assays.The invasion and migration were evaluated by Transwell assays. The protein expression of F-actin, β-catenin,vimentin and N-cadherin was detected by Western blot. Results The degradation rate of Mg-Ag and Mg-Cu was higher than that of pure Mg. The proliferation of Mg-Ag and Mg-Cu groups cells was significantly inhibited;the invasion and migration abilities of Mg-Ag and mg-Cu groups cells were decreased, and the expression of F-actin, β-Catenin, vimentin and N-cadherin was down regulated. Conclusion Mg-Ag and Mg-Cu alloys can regulate EMT of osteosarcoma U2 OS cells through F-actin/β-catenin signaling pathway, and inhibit the proliferation, invasion and migration of U2 OS cells.
