摘  要:为探讨马齿苋总黄酮(portulaca oleracea total flavones, POTF)对昆明小鼠的非酒精性脂肪肝(non-alcoholic fatty liver disease, NAFLD)脂质代谢异常、氧化应激及炎症反应的影响,将小鼠随机分为5组:对照组、NAFLD组、POTF 2.5 mg/kg组、POTF 5 mg/kg组和POTF 10mg/kg组。采用全自动生化分析仪测定丙氨酸氨基转移酶(alanine transaminase, ALT)、天门冬氨酸氨基转移酶(aspartate aminotransferase, AST)、三酰甘油(triglyceride, TG)和空腹血糖;H-E和油红O染色观察肝组织损伤和脂质堆积情况;Western blotting检测脂代谢标志物乙酰辅酶A羧化酶(acetyl CoA carboxylase, ACC)和肉毒碱棕榈酰转移酶1(carnitine palmitoyl transferase 1,CPT-1)的表达;试剂盒检测超氧化物歧化酶(superoxide dismutase, SOD)活性、丙二醛(malondialdehyde, MDA)和谷胱甘肽(glutathione, GSH)水平;ELISA检测肝组织IL-6和IL-10水平;免疫组化(immunohistochemistry, IHC)检测NF-κB p65的表达。结果显示,POTF可明显改善肝组织损伤和脂质堆积情况。与NAFLD组比较,POTF 5 mg/kg组和10 mg/kg组ALT、AST、TG、空腹血糖、ACC、IL-6、MDA及NF-κB p65水平显著降低(P<0.05),而CPT-1水平、GSH水平、SOD活性及IL-10水平显著升高(P<0.05)。该研究提示,POTF可能通过减轻脂质代谢异常、氧化应激和炎症反应改善高脂饮食小鼠NAFLD。This study investigated the effects of portulaca oleracea total flavones(POTF) on lipid metabolism, oxidative stress and inflammatory response in Kunming mice with non-alcoholic fatty liver disease(NAFLD). The mice were randomly divided into 5 groups: control group, NAFLD group, POTF 2.5 mg/kg group, POTF 5 mg/kg group and POTF 10 mg/kg group. Serum alanine transaminase(ALT), aspartate aminotransferase(AST), triglyceride(TG) and fasting blood glucose were determined by the automatic biochemical analyzer. H-E and oil-red O staining were used to observe liver tissue damage and lipid accumulation. Expressions of acetyl CoA carboxylase(ACC) and carnitine palmitoyl transferase 1(CPT-1) were analyzed by Western blotting. The activity of superoxide dismutase(SOD), concentrations of malondialdehyde(MDA) and glutathione(GSH) were measured by the test kits. ELISA was used to detect IL-6 and IL-10 in liver tissues. Immunohistochemistry(IHC) was performed to detect NF-κB p65 expression. The results showed that POTF significantly improved liver tissue damage and lipid accumulation. Compared with the NAFLD group, ALT, AST, TG, fasting glucose, ACC, IL-6, MDA and NF-κB p65 levels in mice treated with 5 and 10 mg/kg of POTF were significantly reduced(P<0.05),while the levels of CPT-1, GSH, SOD activity and IL-10 were significantly increased(P<0.05). It suggests that POTF could improve the NAFLD of high-fat diet mice by reducing abnormal lipid metabolism, oxidative stress and inflammatory response.
