摘 要:目的:通过建立血管性痴呆(VD)大鼠模型,观察电针对VD大鼠学习记忆能力改善和对海马CA1区神经元中沉默信息调节因子1(SIRT1)、过氧化物酶体增殖物激活受体γ辅激活因子1α(PGC1-α)的表达水平的调节作用,探讨中医电针法对VD的神经保护机制。方法:雄性SD大鼠24只,随机分为对照组、模型组和电针组。采用结扎双侧颈总动脉法制作VD大鼠模型,模型制备后,电针组通过电针刺激“百会”穴和“足三里”穴,2Hz、30min连续针刺治疗4周。采用Morris水迷宫法检测各组大鼠的空间学习记忆能力,尼氏染色法比较各组大鼠的海马CA1区细胞形态,免疫组织化学染色法检测大鼠海马CA1区SIRT1及PGC1-α蛋白表达水平的变化。结果:(1)Morris水迷宫实验结果显示:与对照组相比,模型组大鼠在第5天时,平均逃避潜伏期明显升高(P<0.05),在平台象限活动的时间比率显著下降(P<0.05);与模型组相比,电针组大鼠第5天时,平均逃避潜伏期明显下降(P<0.05),在平台象限活动的时间比率显著高于模型组(P<0.05)。(2)尼氏染色发现对照组大鼠海马CA1区神经细胞形态结构完整,核仁清晰可见,尼氏颗粒丰富;模型组海马区CA1区细胞形态多呈空泡状,核仁固缩;电针组海马CA1区的异常形态学变化相对减轻。(3)免疫组织化学实验结果显示:与对照组相比,模型组大鼠海马CA1区SIRT1和PGC1-α阳性神经元数量明显减少(P<0.05),平均灰度值明显增加(P<0.05);与模型组相比,电针组大鼠海马CA1区SIRT1阳性神经元数量明显增加(P<0.05),平均灰度值明显降低(P<0.05)。结论:电针通过通路SIRT1/PGC1-α改善了VD大鼠学习记忆能力,对VD治疗提供一定中医研究基础。Objective:By establishing a rat model of vascular dementia(VD),observe the improvement of VD rats on the learning and memory ability and regulating action of expression level in silent information regulator 1(SIRT1)and peroxisome proliferator-activated receptorγcoactivator 1α(PGC1α)in neurons of the hippocampal CA1 region by electroacupuncturethe,explore the neuroprotective mechanism of traditional Chinese medicine electroacupuncture on vascular dementia.Methods:24 male SD rats were randomly divided into control group,model group and electroacupuncture group.The VD rat model was made by ligating the bilateral common carotid arteries.After the model was prepared,the electroacupuncture group stimulated the“Baihui”and“Zusanli”points by electric acupuncture with 2Hz of the frequency on sparsw wave for 30 minutes,and continued acupuncture for 4 weeks.Morris water maze was used to observe the spatial learning and memory ability of rats.Nissl staining was used to compare the cell morphology of hippocampal CA1 area of rats;immunohistochemical was used to detection the SIRT1 and PGC1-αprotein expression levels hippocampal CA1 area of rats.Results:(1)Results from Morris water maze experiment:compared with the control group,on the 5th day of the model group,the average escape latency was significantly increased(P<0.05),and the time ratio of activities in the platform quadrant was significantly decreased(P<0.05);Compared with the model group,on the 5th day of the electroacupuncture group,the average escape latency was significantly reduced(P<0.05),and the time ratio of activities in the platform quadrant was significantly higher than that of the model group(P<0.05).(2)In Nissl staining,it was found that the morphology and structure of the neurons in the CA1 area of the hippocampus of the control group were complete,with clear nucleoli and abundant Nissl granules;the CA1 area of the hippocampus of the model group was mostly vacuolated with pyknosis of nucleoli.The abnormal morphological changes in CA1 area of elec
