法舒地尔对糖尿病小鼠认知障碍作用的研究

摘  要:目的:研究法舒地尔对糖尿病小鼠认知功能的作用。方法:将小鼠随机分组:正常对照组10只、实验组20只,实验组小鼠造成糖尿病模型小鼠,并随机分为模型组、法舒地尔组,造模成功8周后利用Morris水迷宫系统测评动物认知能力,测试完成后法舒地尔组给予法舒地尔腹腔注射,正常对照组、模型组腹腔注射同等量生理盐水,持续8周,再行水迷宫检测动物认知功能后处死。结果:Morris水迷宫实验显示:(1)寻台时间:造模8周后模型组和法舒地尔组的寻台时间明显长于对照组(P<0.05),但模型组和法舒地尔组无明显差异(P>0.05);三组对应用药8周后,模型组寻台时间(38.80±4.39)s最长,法舒地尔组(33.71±5.88)s次之,对照组(22.50±5.89)s最短,且两两比较均有差异(P<0.05)。(2)目标象限停留时间百分比:造模成功后8周,进行水迷宫显示:模型组和法舒地尔组停留时间显示无差异,但均显著短于对照组(P<0.05),三组对应用药后比较显示:模型组(24.00±3.05)%和法舒地尔组(29.40±7.53)%出现差异(P<0.05),且模型组和对照组差异更显著(P<0.01)。给予法舒地尔治疗前后,进行组内比较,法舒地尔组寻台时间和平台象限时间百分比均有差异(P<0.05),而模型组和对照组腹腔注射生理盐水后组内比较无显著差异。结论:法舒地尔对糖尿病小鼠的认知功能有一定改善作用。Objective:To study the effect of fasudil on cognitive function in diabetic mice.Methods:The mice were randomly divided:10 normal control group and 20 experimental group.The mice in the experimental group were made into diabetic model mice,and they were randomly divided into model group and fasudil group.After 8 weeks of successful modeling,the Morris water maze test was used to evaluate the animal’s cognitive ability.After the test,fasudil group was intraperitoneally injected with fasudil.The normal control group and model group were intraperitoneally injected with the same amount of normal saline for 8 weeks,and then the animals were killed after the cognitive function was detected by water maze.Results:Morris water maze test showed that:(1)The platform seeking time:8 weeks after modeling,the platform seeking time of model group and fasudil group was significantly longer than that of control group(P<0.05),but there was no significant difference between model group and fasudil group(P>0.05).Three groups were given corresponding administration for 8 weeks,the platform seeking time in model group was the longest(38.80±4.39)s,followed by fasudil(33.71±5.88)s,and that in control group was the shortest(22.50±5.89)s(P<0.05).(2)The percentage of residence time in the target quadrant,the water maze showed that there was no difference in the residence time between the model group and the fasudil group,which were significantly shorter than the control group(P<0.05).Three groups were given corresponding administration for 8 weeks,the difference between the model group(24.00±3.05)%and fasudil group(29.40±7.53)%was statistically significant(P<0.05),and the difference between the model group and the control group was more significant(P<0.01).Before and after treatment with fasudil,there were differences in platform seeking time and platform quadrant percentage in fasudil group(P<0.05),but there was no difference between model group and control group after intraperitoneal injection of normal saline(P>0.05).Conclusion:Fas
