摘 要:为开发新型非病毒基因载体,研究了烷基锍类化合物作为基因载体的性能。通过动态光散射实验测定了烷基锍类化合物与质粒DNA复合物的粒径和Zeta电位,通过MTT法研究了烷基锍类化合物的细胞毒性,并通过荧光显微镜观察了复合物的细胞摄取情况。结果表明,4个烷基锍类化合物与质粒DNA在硫磷比(S/P)为40/1、50/1时形成复合物的粒径均在160~210 nm之间;烷基链较短(C12)时,复合物的Zeta电位为负值,烷基链较长(C16)时,复合物的Zeta电位为正值,在+20 mV左右;细胞毒性检测结果显示,在较低浓度下,烷基锍类化合物与PEI相比,细胞毒性无明显差异,在浓度大于6.25μg·mL^(-1)时,烷基锍类化合物的细胞毒性强于PEI;细胞摄取能力检测结果显示,在S/P为40/1时,HepG2细胞存活状态较差,不能有效摄取复合物。该研究为烷基锍类化合物作为基因载体的开发提供了依据。In order to develop new type non-viral gene vectors,we studied the properties of alkyl-sulfonium compounds as gene vectors.In this study,we determined the particle size and Zeta potential of the condensates formed by alkyl-sulfonium compounds and plasmid DNA by dynamic light scattering experiments,studied the cytotoxicity of alkyl-sulfonium compounds by MTT assay,and observed the cellular uptake of the condensates by fluorescence microscopy.The results show that the particle sizes of the condensates range from 160 nm to 210 nm at S/P ratio of 40/1 and 50/1.When the alkyl chain is shorter(C12),the Zeta potential of the condensates is negative,while when the alkyl chain is longer(C16),the Zeta potential is positive,about+20 mV.The results of cytotoxicity assay show that,compared with PEI,the alkyl-sulfonium compounds show similar cytotoxicity at a lower dose,but higher cytotoxicity at a higher dose(greater than 6.25μg·mL^(-1)).The results of cellular uptake capacity test show that HepG2 cells have poor survival status and can not effectively absorb the condensates when the S/P ratio is 40/1.This study provides a basis for the development of alkyl-sulfonium compounds as gene vectors.
