摘 要:目的探讨阻塞性睡眠呼吸暂停大鼠海马区HMGB1、TLR4、NF-κB的表达变化。方法将60只雄性SD大鼠随机分为对照组和模型组(各30只),经咽腔多点注射医用透明质酸钠凝胶制备OSAS大鼠模型。于造模后2、4、6周组通过HE染色观察海马CA1区神经细胞的病理形态,免疫组织化学染色观察海马CA1区HMGB1及TLR4的阳性表达,应用Western blot法检测海马CA1区HMGB1、TLR4、NF-κB蛋白的表达水平。结果HE染色结果显示,与对照组比较,模型组各期海马CA1区神经元排列紊乱,结构稀疏,核仁模糊不清,核固缩、裂解。免疫组化结果显示,实验组各期HMGB1及TLR4的表达较对照组各期均明显增加,差异有统计学意义(P<0.05)。Western blot法检测结果显示,与对照组各期比较,实验2、4、6周组HMGB1、TLR4、NF-κB表达均明显升高,差异有统计学意义(P<0.05)。结论阻塞性睡眠呼吸暂停大鼠海马CA1区神经细胞损伤,并引起该区域HMGB1、TLR4及NF-κB的高表达。Objective To explore the expression changes of HMGB1,TLR4 and NF-κB in the hippocampus of rats with obstructive sleep apnea.Methods Sixty male SD rats were randomly divided into a control group and a model group(30 each),and the OSAS rat model was prepared by multi-point injection of medical sodium hyaluronate gel through the pharyngeal cavity.At 2,4,and 6 weeks after modeling,HE staining was used to observe the pathological morphology of nerve cells in hippocampal CA1 area,immunohistochemi-cal staining was used to observe the positive expression of HMGB1 and TLR4 in hippocampus CA1 area,and Western blot was used to de-tect HMGB1,TLR4,and TLR4 in hippocampus CA1 area.The expression level of NF-κB protein.Results HE staining results showed that compared with the control group,the neurons in the CA1 region of the hippocampus of the model group were arranged disorderly,the structure was sparse,the nucleolus was blurred,and the nucleus was pyknosis and lysis.The immunohistochemical results showed that compared with the control group,the expression was significantly increased in each phase,and the difference was statistically significant(P<0.05).WB results showed that compared with the control group in each phase,the expression of HMGB1,TLR4,and NF-κB in the experimental 2,4,and 6 week groups were all significantly higher,and the difference was statistically significant(P<0.05).Conclusion Nerve cells in the hippocampal CA1 area of obstructive sleep apnea rats were damaged and caused the high expression of HMGB1,TLR4 and NF-κB in this area.
