摘 要:目的:探究氨基酰化酶-1(aminoacylase-1,ACY-1)在儿童神经母细胞瘤组织和细胞中的表达及其意义。方法:采用qRT-PCR和Western blot技术对儿童神经母细胞瘤患者的肿瘤组织和癌旁组织中ACY-1的表达进行检测。然后对人神经母细胞瘤细胞(SH-SY5Y细胞株)进行体外细胞功能实验,运用重组腺病毒载体转染技术过表达或者抑制ACY-1的表达,采用CCK-8法、流式细胞术和Transwell法检测研究ACY-1对该肿瘤细胞增殖、凋亡、迁移和侵袭的影响。结果:qRT-PCR和Western blot实验结果表明,儿童神经母细胞瘤组织中ACY-1的mRNA表达水平和蛋白表达水平均低于癌旁组织。ACY-1过表达不仅可抑制SH-SY5Y细胞株的增殖并诱导其凋亡,而且可抑制其迁移和侵袭;当ACY-1的表达受到抑制时,实验则显示出相反的趋势。结论:ACY-1可能与儿童神经母细胞瘤的发生发展密切相关,它可调节神经母细胞瘤细胞的增殖、凋亡、迁移和侵袭。Objective:To investigate the expression and significance of aminoacylase-1(ACY-1)in the tissues and cells of children with neuroblastoma.Methods:qRT-PCR and Western blot were used to detect the expression of ACY-1 in tumor tissues and adjacent tissues of children with neuroblastoma.Then,in vitro experiments were conducted on human neuroblastoma cells(SH-SY5Y cell line)to overexpress or inhibit the expression of ACY-1 by using recombinant adenovirus vector and transfection technology.The effects of ACY-1 on the proliferation,apoptosis,migration and invasion of the tumor cells were detected by CCK-8 method,flow cytometry and Transwell method.Results:The results of qRT-PCR and Western blot showed that the mRNA and protein expression levels of ACY-1 in tumor tissues were lower than those in the adjacent tissues.ACY-1 overexpression not only inhibited the proliferation of SH-SY5Y cell line and induced its apoptosis,but also inhibited its migration and invasion.Inhibition of the expression of ACY-1 showed the opposite result.Conclusion:ACY-1 may be closely related to the occurrence and development of neuroblastoma in children,and it can regulate the proliferation,apoptosis,migration and invasion of neuroblastoma cells.
