摘 要:目的:检测乳腺癌组织中Chk1(checkpoint kinase1,Chk1)的表达水平,探索其与患者临床病理特征和预后的关系。方法:选取2012年01月至2017年01月在我院病理科存档的乳腺癌组织蜡块115例,应用免疫组织化学法检测其中Chk1蛋白的表达水平。采用卡方检验分析Chk1蛋白的表达情况与临床病理特征的关系;Kaplan-Meier法分析患者生存数据。结果:Chk1蛋白在乳腺癌组织中广泛表达,阳性率为73%;Chk1蛋白的表达水平与患者的年龄、肿瘤的临床分期及p53的表达水平有关(P<0.05);Chk1蛋白的表达与患者5年总生存期(overall survival,OS)有统计学意义(P<0.05);与5年无进展生存期(progress free survival,PFS)、3年OS和PFS无统计学意义(P>0.05)。结论:Chk1在乳腺癌组织中的表达与患者预后相关,表达阴性的患者可能获得时间较长的PFS和OS,表明Chk1有望成为乳腺癌治疗的潜在靶点。Objective:To detect the expression level of checkpoint kinase1(Chk1)in breast cancer tissues and explore its relationship with clinicopathological features and prognosis.Methods:From January 2012 to January 2017,115 cases of breast cancer tissue wax blocks were archived in the department of pathology in our hospital.The expression level of Chk1 was detected by immunohistochemistry.Chi-square test was used to analyze the relationship between the expression of Chk1 and clinicopathological features.Kaplan-Meier method and Log-rank test were used to analyze the survival data of patients.Results:Chk1 was widely expressed in breast cancer tissues with a positive rate of 73%.The expression level of Chk1 was correlated with the age of the patient,the clinical stage of the tumor and the expression level of p53(P<0.05).Kaplan-Meier survival analysis showed that the expression of Chk1 was statistically significant compared with the 5-year OS of the patients(P<0.05).There was no significant difference between the 3-year OS and PFS and 5-year PFS(P>0.05).Conclusion:The expression of Chk1 in breast cancer tissues is related to the prognosis of patients,and patients with negative expression can obtain PFS and OS for a longer time,indicating that Chk1 is expected to become a potential target for the treatment of breast cancer.
