摘  要:稳定、高效的再生体系是进行木纳格葡萄遗传转化的必要条件。本研究对影响不定芽诱导的外植体类型、基本培养基种类、激素组合和暗培养时间进行了测定。结果表明,叶片和叶柄是适合木纳格葡萄组培的外植体,NN69基本培养基适用于不定芽的诱导。叶片和叶柄不定芽诱导的最佳激素组合分别为TDZ(4 mg/L)+IBA(0.2 mg/L)和TDZ(1 mg/L)+IBA(0.2 mg/L),诱导率分别为4.81%和16.30%。在培养初期,3周暗培养明显促进不定芽的生长,且将不定芽转接到1/2MS+IBA(0.1 mg/L)的生根培养基上能够产生不定根,生根率为100%。本研究建立的木纳格葡萄再生体系,可为其后续遗传转化等研究提供技术参考。Stable and efficient regeneration system is the necessary condition for genetic transformation of Vitis vinifera cv.Munage.The types of study,basic medium,hormone combinations and dark culture’s time were determined in this experiment.The results showed that leaves and petioles were suitable explants for tissue culture of grape,NN69 was more suitable for adventitious bud induction and the optimal hormone combination for adventitious bud induction of leaves and petioles were as follows:TDZ(4 mg/L)+IBA(0.2 mg/L)and TDZ(1 mg/L)+IBA(0.2 mg/L),the induction rates were 4.81%and 16.30%respectively.In the early stage of culture,the growth of adventitious buds were promoted obviously when they were cultured 3 weeks in dark condition.And adventitious buds were then transferred to rooting medium of 1/2 MS+IBA(0.1 mg/L),on which could produce adventitious roots,the ratio of root induction was 100%.It was established the regeneration system of Munage grape,which would provide technical reference for genetic transformation.
