摘  要:为研究天麻蛋白在胃蛋白酶水解条件下获得的多肽对铜绿假单胞菌抗菌活性。以牛津杯法测得的抑菌圈大小为指标,考察水解温度、水解时间、水解pH单因素对天麻蛋白水解产物抗菌活性的影响。在单因素基础上设计正交试验获得具最强抑菌活性的多肽水解工艺。结果表明:当水解温度37℃,水解时间3 h和水解pH为4时,天麻蛋白水解物具最强抗铜绿假单胞菌活性。抑菌圈达到17.25 mm。In order to study against Pseudomonas aeruginosa activity of polypeptides which obtained from pepsin hydrolysis of Gastrodiaelata protein,the inhibition zone size measured by Oxford cup method was used as the index,the effects of hydrolysis temperature,hydrolysis time and pH on antibacterial activity of Gastrodiaelata protein hydrolysate were investigated.The orthogonal experiment was designed on the basis of single factor,the optimization of hydrolysis process of polypeptides with the strongest antibacterial activity was obtained from Gastrodiaelata protein hydrolyzed by pepsin.The results showed that the polypeptides hydrolyzed from Gastrodiaelata protein had the strongest activity against Pseudomonas aeruginosa when the hydrolysis temperature was 37℃,the hydrolysis time was 3 h and the hydrolysis pH was 4.The size of inhibition zone reached 17.25 mm.
