摘  要:目的:探讨右美托咪定对缺血性脑损伤大鼠神经功能及认知障碍恢复的影响。方法:将缺血性脑损伤大鼠(n=45)随机平分为三组:模型组、地佐辛组与右美托咪定组。右美托咪定组在再灌注即刻腹腔注射右美托咪定40μg/kg,地佐辛组在再灌注即刻腹腔注射地佐辛40μg/kg,模型组在再灌注即刻腹腔注射相同容量0.9%氯化钠溶液,检测神经功能缺陷评分、血清白介素(IL)-1β、IL-6、micoRNA-329-3p表达水平及海马组织可调控cAMP反应元件结合蛋白(CREB)、白介素-1受体拮抗剂(IL-1RA)蛋白相对表达水平。结果:地佐辛组与右美托咪定组再灌注后第1、3、7天逃避潜伏期、神经功能缺陷评分低于模型组(均P<0.05),右美托咪定组低于地佐辛组(P<0.05);地佐辛组与右美托咪定组再灌注后第7天IL-1β、IL-6表达水平、micoRNA-329-3p相对表达水平低于模型组(均P<0.05),右美托咪定组低于地佐辛组(P<0.05);地佐辛组与右美托咪定组再灌注后第7天的大脑海马组织CREB、IL-1RA蛋白相对表达水平高于模型组(均P<0.05),右美托咪定组高于地佐辛组(P<0.05)。结论:右美托咪定可通过协调micoRNA-329-3p/CREB/IL-1RA轴的表达,促进缺血性脑损伤大鼠神经功能及认知障碍恢复正常,具有很好的脑保护效果。Objective:To investigate the effects of dexmedetomidine on recovery of neurological function and cognitive impairment in rats with ischemic brain injury.Methods:Rats with ischemic brain injury(n=45)were randomly divided into model group,dezocine group and dexmedetomidine group.The dexmedetomidine group was intraperitoneally injected with 40μg/kg dexmedetomidine immediately after reperfusion,and the dezocine group was intraperitoneally injected with 40μg/kg dezocine immediately after reperfusion,and the model group was intraperitoneally injected with 0.9%sodium chloride solution of the same volume immediately after reperfusion.The neurological deficit score,the expression levels of IL-1β,IL-6,microRNA-329-3 p,cyclic-AMP response binding protein(CREB)in and Interleukin-1 receptor antagonist(IL-1 RA)were compared among groups.Results:The escape latency and neurological deficit scores of the dezocine group and dexmedetomidine group were lower than those of the model group at 1,3 and 7 days after reperfusion,and those of the dexmedetomidine group were lower than those of dezocine group(all P<0.05).The serum IL-1βand IL-6 expression levels and the relative expression levels of microRNA-329-3 p at 7 days after reperfusion in the dezocine group and dexmedetomidine group were lower than those in the model group,and those of the dexmedetomidine group were lower than those of the dezocine group(all P<0.05).The relative expression levels of CREB and IL-1 RA proteins in the hippocampus of the dezocine group and the dexmedetomidine group were higher than those of the model group at 7 days after reperfusion,and those of the dexmedetomidine group were higher than those of the dezocine group(all P<0.05).Conclusion:Dexmedetomidine can promote the recovery of neurological function and cognitive impairment in rats with ischemic brain injury by coordinating the expression of microRNA-329-3 p/CREB/IL-1 RA axis,and have good brain protection effect.
