摘 要:目的:分析膀胱尿路上皮癌(BTCC)组织中ERG蛋白表达水平与微血管密度(MVD)和肿瘤分期的相关性。方法:选取62例BTCC患者的癌组织及癌旁组织石蜡标本,检测其ERG蛋白表达、程序性死亡受体配体-1(PD-L1)表达水平以及MVD水平。分析各蛋白阳性表达水平与肿瘤分期的关系,以及ERG、PD-L1蛋白与MVD的相关性。结果:BTCC癌组织中ERG阳性表达39例(62.90%),明显高于癌旁组织中的ERG阳性表达12例(23.08%)(P<0.05);BTCC癌组织中PD-L1阳性表达26例(41.94%),明显高于癌旁组织中的PD-L1阳性表达5例(8.06%)(P<0.05);BTCC癌组织的MVD水平明显高于BTCC癌旁组织(P<0.05)。ERG、PD-L1蛋白阳性表达率、MVD水平在病理类型、肿瘤分期、肿瘤数目中存在统计学差异,在浸润性癌、肿瘤高分期、多发性肿瘤中阳性表达率增高(均P<0.05)。ERG、PD-L1蛋白表达与MVD水平均呈正相关(r=0.426、0.357,P<0.05)。ERG蛋白表达与PD-L1呈正相关(r=0.512,P<0.05)。结论:ERG蛋白在BTCC癌组织中表达异常升高,其可能通过调节PD-L1蛋白表达,参与肿瘤血管的生成和恶性进展。Objective:To analyze the correlation between ERG protein expression level and microvascular density(MVD)and tumor stage in bladder transition cell carcinoma(BTCC).Methods:The paraffin specimens of cancer and adjacent tissues from 62 patients with BTCC were selected to detect the expression of ERG protein,the expression level of programmed death receptor ligand-1(PD-L1)and MVD.The relationship between the positive expression level of each protein and tumor stage,and the correlation between ERG,PD-L1 proteins and MVD were analyzed.Results:The positive expression of ERG in 39 cases(62.90%)of BTCC tissues was significantly higher than that in 12 cases(23.08%)of adjacent tissues(P<0.05).The positive expression of PD-L1 in 26 cases(41.94%)of BTCC tissues was significantly higher than that in 5 cases(8.06%)of adjacent tissues(P<0.05).The MVD level of BTCC tissues was significantly higher than that of adjacent tissues(P<0.05).There were significant differences in the positive expression rates of ERG and PD-L1 proteins,MVD level in pathological type,tumor stage and tumor number,and the positive expression rates increased in invasive cancer,high tumor stage and multiple tumors(all P<0.05).The expressions of ERG and PD-L1 proteins were positively correlated with the level of MVD(r=0.426,0.357,P<0.05).The expression of ERG protein was positively correlated with PD-L1(r=0.512,P<0.05).Conclusion:ERG protein expression is abnormally elevated in BTCC tissues,which may be involved in tumor angiogenesis and malignant progression by regulating PD-L1 protein expression.
