摘 要:目的筛查脑出血大鼠中与Ho-1基因相关的microRNAs。方法将SD大鼠分为脑出血模型组(n=6)与假手术组(n=6)。采用局部注射血栓构建脑出血模型,假手术组大鼠脑部注射生理盐水。HE染色以及神经行为学评分评定模型是否成功建立。检测各组大鼠脑组织HO-1及其相关miRNA(miR-331-3p、miR-298-5p、miR-221-3p、miR-187-5p、miR-7a-5p、miR-3557-3p、miR-673-5p、miR-330-5p、miR-6331、miR-29b-5p、miR-218a-5p、miR-3564-5p、miR-877,miR-31a-5p,miR-455-5p)的表达水平。分离提取两组大鼠脑组织星形胶质细胞后,采用双荧光素酶切报告实验检测miR-877、miR-31a-5p、miR-455-5p与HO-1的靶向关系。结果HE和神经行为学评分结果证明模型建立成功。与假手术组相比,脑出血组大鼠脑组织中HO-1高表达(P<0.01),其相关基因miR-877、miR-31a-5p、miR-455-5p低表达(P<0.01),其余无显著变化。双荧光素酶结果发现,在星形胶质细胞中miR-877与HO-1具有直接靶向关系。结论脑出血大鼠星形胶质细胞中miR-877靶向性调控HO-1,为脑出血损伤的治疗提供了理论基础。Objective To screen HO-1-related microRNAs in cerebral hemorrhage rats.Methods SD rats were divided into cerebral hemorrhage model group(n=6)and sham operation group(n=6).Rats were injected with thrombosis to establish the cerebral hemorrhage model in model group,and rats were injected with saline in sham operation group.HE staining and neurobehavioral score were used to evaluate the successful establishment of model.The expression levels of HO-1 and related miRNAs(miR-331-3p,miR-298-5p,miR-221-3p,miR-187-5p,miR-7a-5p,miR-673-5p,miR-330-5p,miR-6331,miR-29b-5p,miR-218a-5p,miR-3564-5p,miR-877,miR-31a-5p,miR-455-5p)in brain tissues of rats in each group were detected.Then astrocytes were isolated and extracted from brain tissue of rats in each group.The targeting relationships between miR-877,miR-31a-5p,miR-455-5p and HO-1 were detected by dual-luciferase reporter assay.Results HE and neurobehavioral scoring results proved that the model was successfully established.Compared with sham group,HO-1 was highly expressed in the brain tissue of rats in intracerebral hemorrhage group(P<0.01),and its related genes miR-877,miR-31a-5p and miR-455-5p were lowly expressed in the brain tissue of rats(P<0.01).Dual-luciferase reporter assay results showed that miR-877 had direct targeting relationship with HO-1 in astrocytes.Conclusion HO-1 is targetedly regulated by miR-877 in astrocytes of rats with intracerebral hemorrhage,which provides theoretical basis for the treatment of intracerebral hemorrhage injury.
