摘 要:目的利用癌症基因组图谱(TCGA)数据库数据分析肿瘤微环境与卵巢浆液性囊腺癌的临床特征及预后的关系。方法下载TCGA数据库中469例卵巢浆液性囊腺癌患者的临床特征信息及RNA测序数据。通过R软件运用表达谱数据设计的算法(ESTIMATE)计算出肿瘤微环境中免疫细胞评分、间质细胞评分及ESTIMATE评分。利用单因素方差分析统计评分与卵巢浆液性囊腺癌临床病理特征的相关性,利用Kaplan-Meier法分析评分与卵巢浆液性囊腺癌总生存时间的关系。结果免疫细胞评分、间质细胞评分及ESTIMATE评分均与卵巢浆液性囊腺癌患者淋巴结转移相关,但与肿瘤分期、分化程度无关。免疫细胞评分、间质细胞评分及ESTIMATE评分的高低与化疗敏感性、BRCA1/2突变状态以及卵巢浆液性囊腺癌患者的生存预后也无明显关系。结论高免疫细胞评分、间质细胞评分及ESTIMATE评分与卵巢浆液性囊腺癌淋巴结转移明显相关,有助于临床分析卵巢浆液性囊腺癌的进展。Objective To analyze the relationship between tumor micro environment and the clinicopathological features as well as its prognostic value in ovarian serous cystadenocarcinoma patients by using the cancer genome atlas(TCGA)database.Methods Clinical characteristics and RNA sequencing data of 469 patients with ovarian serous cystadenocarcinoma from TCGA database were downloaded.The immune cell score,interstitial cell score and ESTIMATE score in tumor microenvironment were calculated by R software using an algorithm(ESTIMATE)function designed from expression spectrum data.The correlation between the score and the clinicopathological features of ovarian serous cystadenocarcinoma was analyzed by one-way anOVA,and the relationship between the score and the overall survival time of ovarian serous cystadenocarcinoma was analyzed by Kaplan-Meier method.Results Immune,stromal and ESTIMATE score were significantly associated with stage,but did not vary with T stage and grade.Three scores had no relationship with chemotherapy sensitivity and BRCA1/2 mutation.All scores were also not correlated with overall survival rate of ovarian serous cystadenocarcinoma.Conclusion Tumor micro environment is associated with progression of ovarian serous cystadenocarcinoma and can be used as an molecular marker for clinical analysis of ovarian serous cystadenocarcinoma.
