摘 要:目的:探讨miR-518a-5p调控甲状腺鳞癌细胞SW579凋亡的潜在机制。方法:通过实时荧光定量PCR分析33例癌组织和癌旁组织中miR-518a-5p的表达水平。使用miR-518a-5p mimics和miR-518a-5p inhibitor甲状腺鳞癌细胞SW579中过表达或敲低miR-518a-5p,检测细胞的凋亡水平。通过miRDB,软件分析和荧光素酶报告系统分析miR-518a-5p的底物。通过siRNA和质粒分别敲低和过表达底物,检测甲状腺鳞癌细胞SW579的凋亡水平。结果:癌旁组织中miR-518a-5p的相对表达量大于甲状腺鳞癌组织(P<0.05)。过表达miR-518a-5p后,甲状腺鳞癌细胞SW579的凋亡水平上升(P<0.05);敲低miR-518a-5p后,甲状腺鳞癌细胞SW579的凋亡水平下降(P<0.05)。miRDB在线分析发现,miR-518a-5p潜在靶向PRKAR2B,CEP350,PSD3,NUP54,HDAC2,UBE2A,SALL1,PPP1R15B,ELK4,CNOT7。过表达miR-518a-5p后,HDAC2的表达水平下降(P<0.05);敲低miR-518a-5p后,HDAC2的表达水平上升(P<0.05)。miR-518a-5p靶向HDAC2的3端非编码区(P<0.05)。敲低HDAC2后,甲状腺鳞癌细胞SW579的凋亡水平上升(P<0.05)。过表达HDAC2后,甲状腺鳞癌细胞SW579的凋亡水平下降(P<0.05)。同时敲低miR-518a-5p和HDAC2后,甲状腺鳞癌细胞SW579的凋亡水平没有明显变化(P>0.05);同时过表达miR-518a-5p和HDAC2后,甲状腺鳞癌细胞SW579的凋亡水平没有明显变化(P>0.05)。结论:miR-518a-5p通过靶向HDAC2 mRNA的3端编码区降低了HDAC2的表达量,随后促进了甲状腺鳞癌细胞SW579的凋亡。Objective: To explore the potential mechanism of Mir-518 A-5 P regulating the apoptosis of thyroid squamous cell carcinoma cell line SW579. Methods: The expression levels of Mir-518 a-5 p in 33 cancer tissues and adjacent tissues were analyzed by real-time fluorescence quantitative PCR. Mir-518 a-5 p mimics and Mir-518 a-5 p inhibitor THYROID squamous cell cancer cell SW579 were used to overexpress or knock down Mir-518 a-5 p to detect the apoptosis level of the cells. Substrates of Mir-518 A-5 P were analyzed by miRDB, software analysis, and luciferase reporting system. Apoptosis of thyroid squamous cell cell SW579 was detected by siRNA knockdown and plasmid overexpression, respectively.%Results: The relative expression of Mir-518 a-5 p in 114 cases of paracancer tissues was greater than that in 114 cases of thyroid squamous cell carcinoma tissues(P<0.05). After overexpression of Mir-518 a-5 p, the apoptosis level of THYROID squamous cell cell SW579 increased(P<0.05). After mir-518 a-5 p knockdown, the apoptosis level of THYROID squamous cell cell SW579 decreased(P<0.05). Mi RDB online analysis revealed that Mir-518 a-5 P potentially targets PRKAR2 B, CEP350, PSD3, NUP54, HDAC2, UBE2 A, SALL1,PPP1 R15 B, ELK4, CNOT7. After overexpression of Mir-518 a-5 p, the expression level of HDAC2 decreased(P<0.05). After mir-518 a-5 p was knocked down, the expression level of HDAC2 increased(P<0.05). In addition, Mir-518 A-5 P targets the 3-terminal non-coding region of HDAC2(P<0.05). After knocking down HDAC2, the apoptosis level of THYROID squamous cell cell SW579 increased(P<0.05). After overexpression of HDAC2, apoptosis of THYROID squamous cell cell SW579 decreased(P<0.05). When mir-518 a-5 p and HDAC2 were knocked down, the apoptosis level of thyroid squamous cell cell SW579 showed no significant change(P>0.05). After the overexpression of mir-518 a-5 p and HDAC2, the apoptosis level of thyroid squamous cell cell SW579 showed no significant change(P>0.05).
