摘  要:目的本研究拟采用改良Allen’s法制作SCI动物模型,通过检测致伤脊髓组织miR-16的表达探究SCI后细胞过度凋亡的可能机制。方法36只成年SD大鼠随机分为正常组(12只)、假手术组(12只)、模型组(12只),通过改良Allen’s制作SCI模型,观察各组行为学评分(BBB评分)、病理表现及PCR法检测miR-16的表达。结果模型组在术后3 d及7 d的BBB评分与正常组和假手术组对比差异有统计学意义(P<0.01)。模型组中的miR-16表达较正常组和假手术组显著升高(P<0.01)。结论研究认为miR-16参与改良Allen’s法制作大鼠SCI模型后引发神经细胞过度凋亡机制。Objective Establish the modified Allen’s-inducted animal spinal cord injury(SCI)model,to explore the possible mechanism of excessive apoptosis of cell after SCI by detecting the expression of miR-16 in injured spinal cord tissue.Methods 36 healthy adult SD rats were randomly divided into 3 groups:normal control group(12),sham operation group(12)group and model group(12).The SCI animal model was established by modified Allen’s.And then observed the Basso,Beattie&Bresnahan locomotor rating scale(BBB scale),pathological manifestations,and the expression of miR-16 via Real-time PCR.Results The BBB scale of the model group on 3 d and 7 d after operation were statistically significant compared with the normal group and sham operation group(P<0.01).Compared with the normal group and sham operation group,the expression level of miR-16 in model group increased.Conclusion We suggest that MiR-16 is involved in the mechanism of excessive apoptosis of nerve cells in modified Allen’s-induced rat SCI model.
