摘  要:目的:探讨TRIM19在卵巢癌进展中的作用及其分子机制。方法:qPCR检测人卵巢表皮细胞IOSE80、卵巢癌细胞SKOV-3、A2780、OVCAR3中TRIM19表达水平;以siRNA敲减SKOV-3细胞中TRIM19基因表达;以CCK-8方法、Transwell实验、流式细胞术分别检测细胞增殖能力、迁移行为及凋亡率;以RT-qPCR技术检测信号通路分子表达。结果:在卵巢癌细胞SKOV-3、A2780、OVCAR3中,TRIM19表达量较正常表皮细胞IOSE80中明显增强。当TRIM19基因在卵巢癌SKOV-3细胞中被成功敲减后,细胞增殖能力降低74.2%,侵袭能力降低70.0%,凋亡率增加约30倍。在分子水平,敲减TRIM19后,p53、p21、CyclinD、CDK2、Bax基因表达显著增加;同时敲减TRIM19与p53后,p53、p21、CyclinD、CDK2、Bax基因表达略有提高。结论:沉默TRIM19基因抑制卵巢癌细胞增殖、侵袭,诱导凋亡,并且激活p53依赖的凋亡信号,TRIM19促进卵巢癌进程。TRIM19具有成为卵巢癌治疗的新靶点潜力。Objective:To investigate the role and molecular mechanism of TRIM19 in progression of ovarian cancer.Methods:Expression levels of TRIM19 in human ovarian epidermal cells IOSE80,ovarian cancer cells SKOV-3,A2780 and OVCAR3 were detected by qPCR. TRIM19 was knocked down by siRNA. Proliferation ability of SKOV-3 was detected by CCK-8 assay when cell invasiveness by Transwell assay,cell apoptosis by FACs analysis. RT-qPCR method was used to detect expression of critical signaling molecules.Results:Compared with normal IOSE80,expression of TRIM19 in ovarian cancer cells SKOV-3,A2780 and OVCAR3 was significantly increased. After TRIM19 was successfully knocked down,cell proliferation ability was reduced by about 74.2%,cell invasive ability was decreased to 70.0%,but cell apoptosis rate was increased by about 30 folds. In addition,levels of p53,P21,CyclinD,CDK2 and Bax were up-regulated significantly after TRIM19 knockdown in SKOV-3 cells. But the levels of these molecules was increased a little when both TRIM19 and p53 were knocked down in SKOV-3 cells.Conclusion:Silencing of TRIM19 inhibited cell proliferation and invasiveness but induced cell apoptosis in ovarian cancer,and p53-mediated apoptosis signaling was affected.TRIM19 contributed to progression in ovarian cancer. TRIM19 might be a new promising target in therapy of ovarian cancer.
