摘 要:目的掌握云南省动物病毒的多样性和传播风险。方法在云南省师宗县设立10头哨兵牛,定期采血接种细胞进行病毒分离。通过RT-PCR、电镜观察、高通量测序与血清中和试验进行分离病毒的鉴定、全基因组序列获取与流行病学调查。结果2017年从采集的哨兵牛血液样本中分离出2株蓝舌病病毒、3株帕利亚姆病毒、2株流行性出血病病毒和1株待鉴定病毒(YNSZ/V207/2017)。电镜下YNSZ/V207/2017病毒粒子直径约80 nm,呈二十面体对称;全基因组序列分析显示其为哺乳动物正呼肠孤病毒(Mammalian orthoreovirus,MRV)血清1型毒株(MRV-1),病毒的不同基因节段分别与人、白尾鹿、水貂、果子狸和棕蝠上分离MRV的序列相似度最高;血清中和试验表明当地牛、羊和猪中MRV-1的抗体阳性率分别为32.5%、20.0%和42.5%。结论在云南省牛血液标本中分离出1株MRV-1型毒株,该毒株可能为不同宿主来源的基因重配毒株,MRV-1在当地家畜中广泛流行。This study investigated the diversity of viruses and related epidemics in animals in Yunnan Province.Blood samples were collected weekly from ten sentinel cattle in Shizong County,and cells were inoculated for viral isolation.The identity and prevalence of the isolated viruses were confirmed through RT-PCR,electron microscopy observation,whole genome sequencing and serum neutralization testing.Two strains of bluetongue virus,three strains of palyam virus,two strains of epidemic hemorrhagic disease virus and one unidentified virus(YNSZ/V207/2017)were isolated from the blood samples of sentinel cattle.The virion of YNSZ/V207/2017 was approximately 80 nm in diameter and showed icosahedral symmetry under electron microscopy.Whole genome sequence analysis indicated that YNSZ/V207/2017 belonged to mammalian orthoreovirus serotype 1(MRV-1)and shared the highest sequence similarity with the corresponding segments of MRV isolated from humans,Odocoileus virginianus,Mustla vison,Paguma larvata taivana and Eptesicus serotinus.Serum neutralization tests indicated that the positivity rates of MRV-1 antibodies in local cattle,sheep and pigs were 32.5%,20.0%and 42.5%,respectively.In conclusion,an MRV-1 strain isolated from cattle in Yunnan Province may be a gene reassortant strain from different host sources,and the virus was found to be widespread in local livestock.
