摘  要:目的:探讨麻黄碱对卵白蛋白(OVA)诱导的变应性鼻炎大鼠Th2型免疫反应的抑制作用及可能机制。方法:将雌性SD大鼠随机分为4组,对照组、模型组、麻黄碱组(10 mg/kg)和氯雷他定组(2 mg/kg),每组10只。通过OVA诱导变应性鼻炎大鼠模型,并使用药物连续治疗7 d。治疗后,对大鼠进行鼻部症状评分,ELISA法检测大鼠血清免疫球蛋白E(IgE)、IL-4和IL-13水平,苏木精-伊红(HE)染色观察大鼠鼻腔组织形态,qRT-PCR和Western blot检测大鼠鼻黏膜组织TSLP和OX40L的mRNA和蛋白表达,免疫组织化学分析鼻黏膜组织中环氧合酶2(COX2)的表达。结果:与模型组相比,麻黄碱组和氯雷他定组的鼻部症状评分显著降低,鼻腔组织病变明显减轻,COX2阳性细胞率显著降低(P<0.05)。与模型组相比,麻黄碱组和氯雷他定组大鼠血清中IgE、IL-4和IL-13水平显著降低(P<0.05)。与模型组相比,麻黄碱组和氯雷他定组大鼠鼻黏膜组织中TSLP和OX40L的mRNA和蛋白表达水平均显著降低(P<0.05)。结论:麻黄碱对OVA诱导的变应性鼻炎大鼠具有良好的治疗作用,麻黄碱可能通过TSLP/OX40L通路调节变应性鼻炎大鼠Th2型免疫应答,从而发挥治疗作用。Objective:To investigate the inhibitory effect and possible mechanism of ephedrine on Th 2 immune response in rats with allergic rhinitis induced by ovalbumin(OVA).Methods:Female SD rats were randomly divided into 4 groups,control group,model group,ephedrine group(10 mg/kg)and loratadine group(2 mg/kg),each with 10 rats.A rat model of allergic rhinitis was induced by OVA and treated with drugs for 7 consecutive days.After treatment,the nasal symptoms were scored,the levels of serum immunoglobulin E(IgE),IL-4 and IL-13 were detected by ELISA method,the morphology of nasal cavity was observed by hematoxylin-eosin(HE)staining,the mRNA and protein expression of TSLP and OX40L in nasal mucosa were detected by qRT-PCR and Western blot,and the expression of cyclooxygenase 2(COX 2)in nasal mucosa was analyzed by immunohistochemistry.Results:Compared with model group,the nasal symptom scores of ephedrine group and loratadine group were significantly reduced,the nasal cavity tissue lesions were significantly reduced,and the rate of COX 2 positive cells was significantly reduced(P<0.05).Compared with model group,the serum levels of IgE,IL-4 and IL-13 in ephedrine group and loratadine group were significantly reduced(P<0.05).Compared with model group,the mRNA and protein expression levels of TSLP and OX 40 L in nasal mucosa tissue of ephedrine group and loratadine group were significantly reduced(P<0.05).Conclusion:Ephedrine has a good therapeutic effect on OVA-induced allergic rhinitis in rats.Ephedrine may regulate Th 2 immune response of allergic rhinitis rats through TSLP/OX 40 L pathway,thereby exerting a therapeutic effect.
