摘  要:目的探究三七注射液对慢性肾衰竭大鼠肠道菌群多样性结构的影响。方法将120只雄性SD大鼠随机分成正常组、模型组、三七低剂量组、三七中剂量组、三七高剂量组、贝那普利组,每组20只。除正常组外,其他组均采用5/6肾脏切除法建立慢性肾衰竭模型。造模成功后,正常组与模型组大鼠腹腔注射生理盐水10 mL/(kg·d),三七低、中、高剂量组大鼠分别腹腔注射三七注射液1 g生药/(kg·d)、2 g生药/(kg·d)、3 g生药/(kg·d),贝那普利组给予盐酸贝那普利5 mg/(kg·d)灌胃,均1次/d,共8周。末次干预24 h后,腹主动脉采血检测肾功能指标[尿素氮(BUN)、血肌酐(SCr)],收集各组大鼠粪便样本行生物多样性分析(α多样性分析、β多样性分析、OTU分析)。结果正常组与各药物干预组BUN、SCr水平均明显低于模型组(P均<0.05),且三七高剂量组均明显低于三七低剂量组、三七中剂量组和贝那普利组(P均<0.05)。α多样性分析示,模型组Shannon、ACE、Chao指数均明显低于正常组(P均<0.05);三七低、中剂量组ACE、Chao指数均明显高于模型组(P均<0.05),且三七中剂量组明显高于三七低剂量组(P均<0.05);三七高剂量组与贝那普利组Shannon、ACE、Chao指数均明显高于模型组、三七低剂量组、三七中剂量组(P均<0.05),但三七高剂量组均明显低于贝那普利组(P均<0.05)。β多样性分析示正常组与三七低剂量组、三七中剂量组、贝那普利组样本间距离较为接近;在属水平上慢性肾衰竭大鼠的肠道菌群分布比例发生显著改变,三七中剂量组和三七高剂量组中乳杆菌属、毛螺菌属、瘤胃菌属、瘤胃球菌属丰度明显上升,大肠杆菌-志贺菌属丰度明显下降。结论三七注射液可以有效改善慢性肾衰竭大鼠的肠道菌群结构及多样性,保护肾功能。Objective It is to explore the effect of Panax Notoginseng injection on the diversity structure of intestinal flora in rats with chronic renal failure.Methods 120 male SD rats were randomly divided into normal group,model group,Panax notoginseng low-dose group,Panax notoginseng medium-dose group,Panax notoginseng high-dose group,and benazepril group,with 20 rats in each group.Except for the normal group,the other groups were all established chronic renal failure models by 5/6 nephrectomy.After successful modeling,the rats in the normal group and the model group were intraperitoneally injected with normal saline 10 mL/(kg·d),and the rats in the Panax notoginseng low-dose group,medium-dose group and high-dose groups were respectively intraperitoneally injected with Panax notoginseng injection 1 g crude drug/(kg·d),2 g crude drug/(kg·d),3 g crude drug/(kg·d),and the benazepril group was given benazepril hydrochloride 5 mg/(kg·d)by gavage,all once per day,for a total of 8 weeks.24 hours after the last intervention,abdominal aortic blood was collected to detect renal function indicators[urea nitrogen(BUN),serum creatinine(SCr)],and fecal samples of the rats in each group were collected for biodiversity analysis(αdiversity analysis,βdiversity analysis,OTU analysis).Results The levels of BUN and SCr in the normal group and each treatment group were lower than those of the model group(all P<0.05),and the Panax notoginseng high-dose group was significantly lower than the Panax notoginseng low-dose group,the medium-dose group and the benazepril group(all P<0.05).αdiversity analysis showed that the Shannon,ACE and Chao indices of the model group were significantly lower than those of the normal group(all P<0.05);the ACE and Chao indices of the Panax notoginseng low and medium dose groups were significantly higher than those of the model group(all P<0.05),and the medium-dose group was significantly higher than that of the low-dose group(all P<0.05);the Shannon,ACE,and Chao indices in the Panax notoginseng high-dose g
