摘 要:目的观察健脾补肾方对激素性股骨头坏死骨髓间充质干细胞定向分化成骨细胞相关细胞因子的影响,为健脾补肾方治疗激素性股骨头坏死提供相应的分子学基础。方法将健康成年雄性Wistar大鼠随机分为空白组、模型组、健脾补肾方组,模型组及健脾补肾方组建立激素性股骨头坏死模型。造模后健脾补肾方组每天灌胃健脾补肾汤,模型组和空白组每天灌胃等量生理盐水,均连续12周。灌胃结束后,取3组大鼠血清及股骨头组织,分别应用ELISA及Western blot法检测血清及股骨头组织中骨形态发生蛋白2(BMP-2)、转化生长因子-β(TGF-β)、碱性成纤维细胞生长因子(bFGF)、血管内皮生长因子(VEGF)、血小板衍性生长因子(PDGF)、成骨特异性细胞转录因子2(Runx2)表达情况。结果ELISA法检测,健脾补肾方组血清BMP-2、TGF-β、VEGF水平及骨组织中BMP-2水平均明显高于空白组和模型组(P均<0.05),空白组均明显高于模型组(P均<0.05)。Western blot法检测,健脾补肾方组血清BMP-2表达量高于空白组和模型组,血清TGF-β表达量高于模型组。结论健脾补肾方可能通过调控BMP-2、TGF-β而促进骨髓间充质干细胞定向成骨分化,从而防治骨坏死。Objective It is to observe the effect of Jianpi Bushen Decoction on the directional differentiation of bone marrow mesenchymal stem cells into osteocyte-related cytokines in steroid-induced necrosis of the femoral head,and provide the corresponding molecular basis for the treatment of steroid-induced necrosis of the femoral head with Jianpi Bushen Decoction.Methods Healthy adult male Wistar rats were randomly divided into blank group,model group,Jianpi Bushen Decoction group,the steroid-induced femoral head necrosis models were established in the model group and Jianpi Bushen Decoction group.After modeling,the Jianpi Bushen Decoction group was daily given Jianpi Bushen Decoction by gavage,and the model group and the blank group were daily given the same amount of normal saline,all were continuously treated for 12 weeks.At the end of treatment,the serum and femoral head tissue of the rats in the three groups were collected,and the expression of bone morphogenetic protein 2(BMP-2),transforming growth factor-β(TGF-β),basic fibroblast growth factor(bFGF),vascular endothelial growth factor(VEGF),platelet-derived growth factor(PDGF),osteoblast-specific cell transcription factor 2(Runx2)in serum and femoral head tissue were respectively detected by ELISA and Western blot method.Results ELISA method showed that the levels of BMP-2,TGF-β,VEGF in serum and BMP-2 in bone tissue in Jianpi Bushen Decoction group were significantly higher than those in the blank group and model group(all P<0.05),and the blank group was significantly higher than the model group(all P<0.05).Western blot assay showed that the expression of serum BMP-2 in Jianpi Bushen Decoction group was higher than that in the blank group and model group,and the expression of serum TGF-βwas higher than that in the model group.Conclusion Jianpi Bushen Decoction may promote the directional osteogenic differentiation of bone marrow mesenchymal stem cells by regulating BMP-2 and TGF-β,thereby preventing and treating osteonecrosis.
