摘  要:为了研究日粮中添加不同水平的棉籽糖对肉鸡肠道菌群的影响,试验将144只健康的14日龄白羽肉鸡随机分成4组,每组6个重复,每个重复6只。对照组饲喂基础日粮,处理组T1、T2、T3分别在基础日粮中添加0.1%、0.2%和0.3%的棉籽糖,试验期为28 d。试验期间每天记录各组采食量,试验第1天和第28天每只鸡空腹称重,计算平均日采食量、平均日增重和料重比。于肉鸡42日龄时,每组选取3只鸡屠宰,采集各组盲肠内容物样品进行16S rRNA测序,根据测序结果进行微生物群落操作分类单元(OTUs)聚类、等级丰度曲线、稀释曲线、Alpha和Beta多样性以及微生物群落门和属组成及相对丰度分析及微生物功能预测。结果表明:T3组肉鸡料重比显著低于对照组、T1和T2组(P<0.05)。等级丰度曲线和稀释曲线表明测序量足以反映样本的物种多样性以及测序深度。T2和T3组Shannon指数显著高于对照组和T1组(P<0.05),T1、T2和T3组的Chao1指数显著高于对照组(P<0.05)。各组样本能较易的区分开来,且组内的3个样本聚集在一起。厚壁菌门(Firmicutes)、拟杆菌门(Bacteroidetes)、放线菌门(Actinobacteria)、变形菌门(Proteobacteria)、蓝细菌门(Cyanobacteria)和脱铁杆菌门(Deferribacteres)是优势菌门_T1组拟杆菌门相对丰度较其他组低,厚壁菌门相对丰度较其他组高_T2组厚壁菌门、放线菌门、变形菌门、蓝细菌门、脱铁杆菌门相对丰度低于对照组,拟杆菌门相对丰度高于对照组_T3组厚壁菌门、放线菌门、变形菌门和脱铁杆菌门相对丰度低于对照组,拟杆菌门、蓝细菌门相对丰度高于对照组。拟杆菌属(Bacteroides)、乳杆菌属(Lactobacillus)、未定义-拟杆菌属(unclassified_acteroidales)、克里斯滕氏菌科_-7_(Christensenellaceae_-7_roup)等是优势菌属_T1组未分类_胃菌科属(unclassified_uminococcaceae)相对丰度显著高于T2组(P<0.05),拟杆菌属(Bacteroides)和未定义_杆菌In order to investigate the effect of adding different doses of raffinose in diet on intestinal flora of broilers,a total of 144 male 14-day-old white feather broilers were randomly divided into four groups with six replicates per group and six chickens in each group.Broilers in control group were fed a basal diet,and those in treatment groups T1,T2,T3 were fed the basal diet supplemented with 0.1%,0.2%and 0.3%raffinose,respectively.The study was lasted to 28 d.During the experiment,the daily feed intake of each group was recorded.On the 1 st and 28 th day of the experiment,each chicken was weighed on an empty stomach.the average daily feed intake,average daily gain and feed weight ratio were calculated.At 42 days of age,three chickens per group were selected for slaughter,and the cecal contents samples of each group were collected for 16 S rRNA sequencing.The sequencing results were used for microbial community OTUs,rank-abundance curve and rarefaction curve,Alpha and Beta diversity,microbial community phyla and genus composition,relative abundance analysis,and microbial function prediction.The results showed as the feed weight ratio of broilers in T3 group was significantly lower than that in control group,T1 and T2 groups(P<0.05).The rank-abundance curve and rarefaction curve showed that the sequencing amount was sufficient to reflect the species diversity and sequencing depth of the samples.Shannon index in T2 and T3 groups were significantly higher than that in the control group and T1 group(P<0.05).Chao1 index in T1,T2,and T3 groups were significantly higher than those in the control group(P<0.05),and the samples between the treatment groups could be easily distinguished,and the three samples in the group are clustered together.Firmicutes,Bacteroidetes,Actinobacteria,Proteobacteria,Cyanobacteria,and Deferribacteres were the dominant phylum.Compared to other groups,the relative abundance of Bacteroidetes was lowest and Firmicutes was highest in T1 group.The relative abundance of Firmicutes,Actinobacteria,Pro
