摘 要:目的:探讨肝癌衍生生长因子(HDGF)的小分子干扰RNA(siRNA)对BALB/c小鼠结直肠癌肝转移的影响。方法:选择78只SPF级BALB/c雄性小鼠,应用盲肠造疝原位接种瘤块法建立小鼠结直肠癌肝转移模型。将成功建模的小鼠随机分为3组,即HDGF-siRNA组、NS-siRNA组与生理盐水(NS)组。根据分组情况,分别于肿瘤原位注射HDGF-siRNA、非特异性siRNA(NS-siRNA)及生理盐水,剂量均为0.15 ng/kg,连续治疗30 d。分时间点记录各组小鼠体质量的变化。治疗结束3 d后处死小鼠,肉眼及肝脏切片苏木素-伊红(HE)染色观察各组结直肠癌肝转移情况。结果:治疗5、10、15、20、25和30 d后,HDGF-siRNA组的体质量均明显高于NS-siRNA组与NS组,NS-siRNA组的体质量均明显高于NS组(P<0.05)。HDGF-siRNA组的肝转移结节数明显少于NS-siRNA组及NS组,肝转移率明显低于NS-siRNA组及NS组(P<0.05)。NS-siRNA组的肝转移结节数明显少于NS组(P<0.05),NS-siRNA组与NS组的肝转移率比较,差异无统计学意义(P>0.05)。结论:HDGF-siRNA可靶向抑制HDGF,从而降低小鼠结直肠癌肝转移的发生,HDGF在结直肠癌肝转移中可能起了重要作用。Objective:To investigate the effect of small interfering RNA(siRNA)of hepatoma-derived growth factor(HDGF)on liver metastasis of colorectal carcinoma in BALB/c mice.Method:A total of 78 SPF BALB/cmale mice were used to establish the model of hepatic metastasis of colorectal cancer.The tumor-bearing mice were randomly divided into three groups,namely HDGF-siRNA group,NS-siRNA group and normal saline(NS)group.According to the grouping,HDGF-siRNA,normal saline nonspecific siRNA(NS-siRNA)and normal saline were injected into the tumor in situ at the dose of 0.15 ng/kg for 30 d.The changes of body weight of each group were recorded at time points.The mice were killed 3 d after treatment.The liver metastasis of colorectal cancer in each group was observed by naked eye and hematoxylin-eosin(HE)staining.Result:After treatment for 5,10,15,20,25 and 30 d,the body weights of HDGF-siRNA group were significantly higher than those of NS-siRNA group and NS group(P<0.05).The number of liver metastasis nodules in HDGF-siRNA group was significantly less than those in NS-siRNA group and NS group,and the liver metastasis rate was significantly less than those in NS-siRNA group and NS group(P<0.05).The number of hepatic metastatic nodules in NS-siRNA group was significantly less than that in NS group(P<0.05),there was no significant difference in liver metastasis rate between NS-siRNA group and NS group(P>0.05).Conclusion:HDGF-siRNA can inhibit HDGF and reduce the occurrence of liver metastasis of colorectal cancer in mice,HDGF-siRNA may play an important role in liver metastasis of colorectal cancer.
