摘  要:以荸荠为试材,采用真空充氮包装鲜切荸荠,探究真空充氮包装对鲜切荸荠的护色效果及其内在机制。研究结果表明,真空充氮包装能显著抑制鲜切荸荠在贮藏过程中a^(*)值、b^(*)值上升和L^(*)值的下降;酶活性分析发现真空充氮包装能显著抑制鲜切荸荠贮藏过程中苯丙氨酸解氨酶(PAL)活力的上升,而多酚氧化酶(PPO)活性变化与荸荠黄化的相关性不大。荧光定量PCR(qRT-PCR)分析表明,真空充氮包装能有效抑制黄酮合成关键结构基因CwPAL、CwC4H、CwCHS和CwF3H的表达,同时显著抑制黄酮合成相关转录因子CwMYB12的表达。以上结果表明,真空充氮包装能够抑制黄酮合成相关酶活性和基因表达,从而有效控制荸荠黄化。Using Chinese water chestnut as the test material,nitrogen-filled vacuum packaging of fresh-cut Chinese water chestnuts was used to investigate effect and mechanism of vacuum nitrogen-filled packaging on color protection of fresh-cut Chinese water chestnut.The result indicated that nitrogen-filled vacuum packaging significantly inhibited the increase of a^(*),b^(*)value and the decrease of L^(*)value during storage.The enzyme activity analysis showed that nitrogen-filled vacuum packaging remarkable inhibited the increase of phenylalanine ammonia-lyase (PAL) activity during storage,and the activity of polyphenol oxidase (PPO) showed no direct correlation with the yellowing of Chinese water chestnuts.The quantitative Real-time PCR (qRT-PCR) analysis showed that nitrogen-filled vacuum packaging effectively inhibited the expression of key structural genes in flavonoid synthesis pathway,including CwPAL,CwC4H,CwCHS and CwF3H,and suppressed the expression of CwMYB12 which was a transcription factor related to flavonoid synthesis.In conclusion,the delayed yellowing of Chinese water chestnuts maybe attribute to the inhibition effect of nitrogen-filled vacuum packaging on activity of the key enzyme and gene expression related to flavonoid synthesis.
