摘 要:现有的右旋糖酐原料药生产工艺中,用乙醇沉淀法将右旋糖酐发酵液中的高分子右旋糖酐分离为粗酐,不经干燥直接溶解为右旋糖酐溶液,盐酸水解分离纯化为药用右旋糖酐,水解液中有乙醇残留。因此用右旋糖酐酶替代盐酸水解右旋糖酐时,必须考虑乙醇对右旋糖酐酶作用的影响。进行了乙醇对右旋糖酐酶酶解反应、稳定性及右旋糖酐酶解的影响的研究。结果表明,在一定条件下,右旋糖酐溶液中乙醇含量30.0%以下对右旋糖酐酶酶解反应本身无影响。低温(4℃)条件下,右旋糖酐酶在30%以下的乙醇溶液中相对稳定。在热的乙醇溶液(50℃)中造成酶蛋白凝聚变性而失活,乙醇浓度越大,这种现象越严重,不同的乙醇浓度,右旋糖酐酶活力损失不同。乙醇对右旋糖酐酶解的影响因乙醇浓度的不同而不同,5.0%以下影响较小,7.5%~15.0%影响在同一水平,17.5%以上影响加剧,但是乙醇对右旋糖酐酶解的影响是可控的。In the current production process of dextran API,high molecular dextran in dextran fermentation broth was separated into crude dextran by ethanol precipitation method,and then was dissolved into dextran solution without drying,and then hydrolyzed and purified into medicinal drdextran by hydrochloric acid hydrolysis.There was residual ethanol in the hydrolysate.Therefore,when dextranase was used to replace hydrochloric acid hydrolyzes dextran,the effect of ethanol on dextranase must be considered.The effect of ethanol on the enzymatic hydrolysis reaction,stability and enzymatic hydrolysis of dextranase was studied.The results showed that under certain conditions,the content of ethanol in dextran solution below 30.0%had no effect on the enzymatic hydrolysis of dextranase.At low temperature(4℃),dextranase was relatively stable in ethanol solution with less than 30%ethanol.The enzyme protein was denatured and inactivated in hot ethanol solution(50℃).The higher the concentration of ethanol,the more serious this phenomenon was.At different ethanol concentrations,the loss of dextranase activity was different.The effect of ethanol on dextran enzymatic hydrolysis varied with the concentration of ethanol.The effect of less than 5.0%on enzymatic hydrolysis of dextran was small,7.5%~15.0%was at the same level,and the effect of more than 17.5%was intensified.However,the effect of ethanol on dextranase hydrolysis could be controlled.
