摘 要:目的观察二甲双胍(metformin)对1-甲基-4-苯基吡啶离子(1-methyl-4-phenylpyridine ion,MPP;)诱导损伤的神经细胞SH-sy5y的保护作用,探索二甲双胍治疗帕金森病(Parkinson’s disease,PD)的作用机理。方法采用(MPP;)诱导PD细胞模型24h,分组为Control组、MPP;组(800mol)、Metformin组(二甲双胍预保护2h,1mM),以CCK-8检查细胞的存活率,检测乳酸脱氢酶(lactate dehydrogenase,LDH)释放情况,细胞免疫荧光观察细胞形态,流式仪检测细胞凋亡情况,Western blot检测各组细胞凋亡相关蛋白Bcl-2,Bax表达以及自噬相关蛋白LC3,p62表达情况。结果与MPP+组相比,Metformin组可促进细胞的存活,减少细胞膜破坏,与MPP;组相比,Metformin组Bcl-2水平升高,Bax表达下降,同时LC3含量升高,p62表达下降。结论二甲双胍能够减轻MPP;诱导的PD模型细胞的损伤,其机制可能与诱导自噬及抑制细胞凋亡有关。Objective To observe the protective effect of metformin on 1-methyl-4-phenylpyridine ion(MPP;)-induced injury of nerve cells SH-sy5 y,and to explore the mechanism of metformin in treating Parkinson’s disease(PD).Methods The PD cell model was induced by(MPP;)for 24 h.Cells were divided into the control group,MPP+group and Metformin group(metformin pre-protection for 2 h plus MPP;).The survival rate of cells was checked by CCK-8,and the release of lactate dehydrogenase(LDH)were detected.Cell morphology was observed by immunofluorescence,cell apoptosis was detected by flow cytometry and the expression of Bcl-2 and Bax in each group and the expression of autophagy-related proteins LC3 and p62 were checked by Western blot.Results Compared with the MPP;group,the Metformin group increased the expression of Bcl-2 and decreased the expression of Bax,while the expression of LC3 increased and the expression of p62 decreased.Conclusion Metformin can reduce the damage of PD model cells induced by MPP;,and its mechanism may be related to the induction of autophagy and inhibition of cell apoptosis.
