摘 要:选取1日龄健康雌性黄羽肉鸡360只,随机分成4个处理组,每个处理6个重复,每个重复15只鸡。4个处理组分别饲喂新鲜大豆油饲粮(Ⅰ组,对照组)、新鲜大豆油+葛根素(750 mg/kg)饲粮(Ⅱ组)、氧化大豆油饲粮(Ⅲ组)和氧化大豆油+葛根素(750 mg/kg)饲粮(Ⅳ组)。在28,56日龄时,每个重复随机选取1只鸡,断颈处死,取胸腺、脾脏和法氏囊备用。结果显示,与对照组相比,饲喂含氧化大豆油饲粮显著提高28日龄肉鸡胸腺、法氏囊和56日龄肉鸡脾脏丙二醛(MDA)含量(P<0.05),显著降低28日龄肉鸡脾脏及56日龄肉鸡胸腺和法氏囊总抗氧化能力(T-AOC),并显著降低28日龄肉鸡法氏囊、脾脏超氧化物歧化酶(SOD)活性(P<0.05)。与对照组相比,饲喂含氧化大豆油饲粮显著降低28,56日龄肉鸡胸腺血红素氧合酶-1(HO-1)和法氏囊中核因子E2相关因子2(Nrf2)、醌氧化还原酶1(NQO1)mRNA表达(P<0.05),显著降低28日龄肉鸡胸腺和法氏囊SOD以及脾脏Nrf2、NQO1 mRNA表达(P<0.01),且显著降低56日龄肉鸡胸腺Keap1、Nrf2和脾脏HO-1 mRNA表达(P<0.01)。与饲喂氧化大豆油饲粮组相比,饲粮中添加葛根素显著降低饲喂氧化大豆油28,56日龄肉鸡胸腺及56日龄脾脏MDA含量(P<0.01),显著提高饲喂氧化大豆油28,56日龄肉鸡脾脏T-AOC和SOD活力以及28日龄胸腺T-AOC(P<0.05),且显著提高饲喂氧化大豆油28,56日龄肉鸡胸腺Keap1、HO-1以及法氏囊SOD mRNA表达(P<0.01)。饲粮中添加葛根素显著提高饲喂氧化大豆油28日龄肉鸡法氏囊Nrf2、Keap1、HO-1、NQO1以及56日龄胸腺SOD和脾脏Keap1、HO-1 mRNA表达(P<0.01)。结果表明,饲喂氧化大豆油降低黄羽肉鸡免疫器官的抗氧化能力,而饲粮中添加葛根素可缓解饲喂氧化大豆油条件下黄羽肉鸡免疫器官抗氧化性能的降低。A total of 360 one-day-old female broilers were randomly divided into four treatment groups with six replicates per treatment and 15 birds per replicate.The broilers in the four treatment groups were given one of four diets:basal diets with fresh soybean oil(control group),basal diets with fresh soybean oil and supplementation of 750 mg/kg puerarin,basal diets with oxidized soybean oil,and diets with oxidized soybean oil and supplementation of 750 mg/kg puerarin.At 28 and 56 d of age,one bird from each replicate was randomly selected and slaughtered for immune organs sample collection.The results showed that compared with the control group,consumption of oxidized oil significantly increased the content of malondialdehyde(MDA)in thymus and bursa of fabricius on 28 d as well as in spleen on 56 d(P<0.05).The total antioxidant capacity(T-AOC)in spleen on 28 dand thymus and bursa of fabricius on 56 d,and the activity of superoxide dismutase(SOD)in thymus,spleen and bursa of fabricius on 28 dwere significantly decreased by consumption of oxidized oil when compared with control group(P<0.05).Consumption of oxidized oil significantly decreased(P<0.05)the mRNA expression levels of hemeoxygenase-1(HO-1)in thymus,nuclear factor erythroid-2 related factor 2(Nrf2)and NADPH∶quinone oxidoreductase-1(NQO1)in bursa of fabricius on both 28,56 d,SOD in thymus and bursa of fabricius,and Nrf2 and NQO1 in spleen on 28 d,as well as Kelch-like ECH2 associated protein 1(Keap1)and Nrf2 in thymus on 56 das compared to control group.Dietary supplementation of puerarin significantly decreased MDA content in thymus on both 28,56 dand spleen on 56 d(P<0.01),and significantly enhanced the T-AOC and SOD activity in spleen on both 28,56 das well as T-AOC in thymus on28 dof broilers fed with oxidized oil as compared to those of broilers fed with oxidized oil without pueratin supplementation(P<0.05).The mRNA expression levels of Keap1 and HO-1 in thymus,and SOD in bursa of fabricius of broilers fed with oxidized oil on both 28 dand 56 dwere also
